To evaluate the biosafety of transgenic Bacillus Thuringiensis (Bt) rice (followed as Bt rice) during the storage, a serial of studies on Bt rice expressing Cry1Ac/ Cry1Ab gene and its parent rice (control) were conducted in the warehouse of Insect Ecology Laboratory of Hua Zhong Agricultural University. First, studied on the effects of transgenic Bt rice on the storage pests in the reposition. Then compared the enzyme activity of warehouse harmful insects. At the same time, separation and identification of Aspergillus, and then infest the transgenic Bt rice. The results were as follows.In the storage, the Bt rice had influence on the growth and development of Sitophilus zeamais, but had no negative effect on the Tribolium castaneum. In all the treatments, the survival rate of Sitophilus zeamais in the Bt rice had significant higher than control, as for the Tribolium castaneum, between the Bt rice and it's control had no significant difference.According to the analysis of enzyme activity, the results indicated that besides the CarE of Sitophilus zeamais in SY non Bt rice had higher than in SY Bt rice and the SOD of Tribolium castaneum in SY non Bt rice had higher than in SY Bt rice, in the other treatments, the CarE, TchE, ADH, SOD, all the enzyme activities in Bt rice had higher than control.4 kinds of pathogenic fungi was isolated from moldy rice, observe the configuration and through the ribosome DNA-ITS sequence analysis, the results showed that all the 4 kinds of pathogenic fungi were the Aspergillus flavus. Inoculation effect was studied with integrated, cut embryo kernels of Bt rice varities and non Bt rice varities inoculated with A. flavus race at different spore concentration, the results revealed that there was significant effect on varieties of rice as well as the kernels with different damaged degree; Significant effect was observed among the spore concentraction of A. flavus also.
