The production of PSP is from Cov-I strain of Chinese Yunzhi (Coriolus versicolor). with the deep-layer submerged of the mycelia as raw material extracted by traditional technology of extraction by water and precipitation by alcohol. PSP can obviously strengthening the immunological function of the mechanism of our body and preventing of cancer. It also has been passed FAD warrant to use as a new biological response modifier substance drug for cancer. The yield of PSP by the popular method, which main process is with the deep-layer submerged of the mycelia as raw material extracted by traditional technology of extraction by water and precipitation by alcohol, is very low. In order to reduce costs,improve yields and revenue, the following three aspects, screening good mutant strain,developing submerged fermentation and extraction technology has been studied in this paper.By means of UV mutation combined with melting wall enzymes tolerance screening, a mutant strain YUBV93 was screened from 301 mutant strains .The strain was well grown in PDA solid-state medium.It produces the amount of mycelia and PSP slightly higher than the original strain did when grown in liquid state medium.By studying extracting technology of invivo polysaccharide peptide of Coriolus versicolor, the optimal extracting technology is confirmed. The total sugal and peptide content of invitro polysaccharide peptide of Coriolus versicolor also has been tested. The result , the yield of PSP and the contents of total sugal and peptide are 4.9%, 36.8% and 26.5% respectively, by the water extraction method;The yield of invivo polysaccharide peptide of Coriolus versicolor and the contents of total sugal and peptide are 10.4%, 39.0%and 28.0% respectively, by the alkali extraction method; The yield of invivo polysaccharide peptide of Coriolus versicolor and the contents of total sugal and peptide are are 3.6%, 37.3% and 27.0% respectively, by the cold alkali extraction method; The yield of invivo polysaccharide peptide of Coriolus versicolor and the contents of total sugal and peptide are 3.8%, 38.2% and 27.8% respectively, by the hot alkali extraction method; The yield of invtro polysaccharide peptide of Coriolus versicolor and the contents of total sugal and peptide are 1.15%, 50.2% and 30.5% respectively, by the water extraction method.The optimal extract condition of PSP has been optimized;first use traditional technology of extraction by water and precipitation by alcohol to extract PSP then use cold alkali and hot alkali extract twice and precipitation by alcohol to get invivo polysaccharide peptide of Coriolus versicolor.In the research of mycelium fermentation process, it is found that Mg2+,Ca2+,EDTA,glycerol and vegetable oils can increase the yield of deep-layer submerged of the mycelia. The best concentration of Mg2+,Ca2+,EDTA,glycerol and vegetable oils is 0.3%,0.3%,0.0002%,3%,7% respectively, under this condition the maximal dry mycelium weight reached 1.52g/150mL,1.41g/150mL,1.40g/150mL,1.69g/150mL,2.25g/150mL and the output of mycelium has increased by 26%,17%,18%,37%,87% respectively. But the yield of psp from the mycelia and the content of total sugal has no obvious change, they are 4.93%,4.87%,4.92%,4.97%,4.99% and 36.97%,37.02%,36.85%,36.84%,36.90% respectively.We developed the production process.firstly,use YUBV93 for the deep-layer submerged of the mycelia,secondly,add 7% vegetable oils and 0.3% MgCl2 to fermentation medium,lastly,secelt the optimal extracting technology. under this condition the maximal dry mycelium weight reached 3.40g/L,The yield of invivo polysaccharide peptide of Coriolus versicolor and the contents of total sugal and peptide are 12.52%, 37.85%and 28.13% respectively.Compare with the traditional production process, the maximal dry mycelium weight,The yield of invivo polysaccharide peptide of Coriolus versicolor and the contents of total sugal and peptide are increase by 86.8%,153.9%,2.5% and 6.1% respectively.
