Takifugu Rubripes is a member of genus pufferfish, family Tetraodontidae, order Tetraodontiformes, class Osteichthyes, often better known by the name fugu. Fugu is one of the most important economical fish in marine aquaculture. It has particular delicious and economic values.Genome sequencing projects of fugu that has been incorporated in"Human Genome Project" is the prototype for the Human Genome,furthermore, whole genome sequencing was completed in 2002. Asadvances in the research of fugu genome, it is necessary to study ongrowth and development, pathology, and flesh development/quality.However, studies of the fugu are limited to gene evolution andenviomment toxicology. So proteomic research of fugu has enormousmeanings and few proteomic research has been reported. In this paper, westudied fugu liver and kidney at the protein level to set up a system forfugu proteome, and a better resolution 2-DE map was established.1. In fugu liver protein profile, 66 proteins were identified by proteomicsand 26 proteins were firstly found in fugu. The novel proteins involvedin metabolism were retinol dehydrogenase family andacyl-CoA-binding domain-containing protein 5A; ones related togenetic information processes were tetratricopeptide repeat domain 5and TAF5-like RNA polymerase II; ones related to development, suchas SH3-binding domain protein, cyclin D1, hyaluronan proteoglycanlink protein, Bonnie and clyde, tuba1 protein, Xin actin binding repeatcontaining protein, type I cytokeratin, enveloping layer and forminhomology; ones involved in celluar processes were chaperonincontaining TCP1, CTAGE family, coatomer protein complex, nuclear envelope containing spectrin repeat, kinesin family member 5B and mitogen-activated protein kinase.2. In fugu kidney protein profile, 38 proteins were identified by proteomics and 19 proteins were firstly found in fugu. The novel proteins involved in metabolism are arginyl-tRNA synthetase and H~+ transporting ATP synthase; one related to genetic information processes was polymerase (DNA-directed) delta interacting protein; ones related to development were flavoprotein oxidoreductase and MID1 interacting protein; ones involved in celluar processes were phosphatase and actin regulator, exocyst complex, calponin, keratin and annexin.3. The structural and functional motif of SN4TDR were analyzed with HCA, PROSITE and Motif scan software as a model of bioinformatical analysis. It was discovered that SN4TDR gene was 87624bp in length and comprised 24 exons and 23 introns. SN4TDR showed five main functional domains that included four N-terminal SN-like domains and a C-treminal domains overlapping with SN-like and Tudor domains. SNc1 and SNc2 of the protein could bind to DNA, while SNc3 and SNc4 formed a basic concav surface of the crescent-shaped structure to capture RNA substrates. These results suggested that the protein may play a crucial role in transcriptional and posttranscriptional regulation.
