Girardinia suborbiculata, which belongs to Urticaceae, a plant of annual herb or perennial herb or half-shrub. Girardinia suborbiculata is the main source of nettle in civilian.It is bitter, acrid in taste and cool-natured, slightly poisonous, eliminating wind, to eliminate expectoration, damp elimination, deintoxication, it was used for treat infantile convulsion, aneos, et al. Girardinia suborbiculata is generally used for rheumarthritis, arthritis deformans, et al. Girardinia suborbiculata is wide-ranging distribute in torrid zone Asia and Africa, and it is main distribute in wide-ranging zone of northern latitude 22°35'~40°50', east longitude 80°25'~125°40 in china. Girardinia suborbiculata have a very abundant natural resources, it has high value of medicine and economy. In the recent years, internal scholars pay close attention to value of Girardinia suborbiculata in anti-inflammatory, anti-rheumatic disease, anti-coagulant day by day. The medicine made from Nettle had been on sale in Europe. As yet, Girardinia suborbiculata has not deeply research in internal or overseas. To find out Pharmacological effect and component of anti-bacterial activity of Girardinia suborbiculata, this research research Girardinia suborbiculata C.J.Chen obtained in ChongQing, and using modern science technique in studying on Pharmacological effect and activity component. The finding will provide research data in Pharmacological effect, chemical composition, clinical application for further use of Girardinia suborbiculata.Main research as follow:1. Pharmacological effect of Girardinia suborbiculata C.J.Chen.(1) anti-inflammatory action:Anti-inflammation effect was studied with acute ear edema induced with dimethylbenzene on mice, and carrageenan to cause paw of mice swell. All the high dose group (HDG) and low dose group (LDG) (the dose of HDG and LDG is 2.5g/kg and 1.25g/kg, respectively) can degrade contents of inflammatory mediator as PGE2, and correlate of inflammatory mediator as MDA. And it can increase contents of SOD for anti-inflammatory.(2) On central nervous system.Autonomic activity test show:Autonomic activity frequency of HDG obviously decrease after 3d, and LDG obviously decrease after 4d too, then Girardinia suborbiculata C.J.Chen have conscious-sedation effect.Rotating rods method show:The fall rate of mices fed with HDG or LDG both significantly increased, and it demonstrate that Girardinia suborbiculata C.J.Chen has a strong influence on central nervous system of mice.Elevated plus-maze test show:Compared with the control group, OE% and OT% of mices fed with HDG or LDG decreased, but the difference was not obvious, so Girardinia suborbiculata C.J.Chen has no antianxiety activity.(3) Effect on phagocytosis of mice macrophageEffect on phagocytosis of mice macrophage show:The ethanol extract from Girardinia suborbiculata C.J.Chen can significantly enhanced phagocytic neutral red function of macrophage. And with a dose-effect relationship, so it showed that Girardinia suborbiculata C.J.Chen may have immune-enhancing effect.(4) Effect on anti-analgesic:Hyperuricacidemia model test show that the mices of Hyperuricacidemia model established with 10% xanthine fed with HDG or LDG, the contents of blood uric acid significantly decreased compared with the model group. Girardinia suborbiculata C.J.Chen have anti-gout effect.(5) Effect on antibacterial activityThe ethanol extract from Girardinia suborbiculata C.J.Chen has antibacterial activity on S.aureus MR, E.coli, P.vulgaris, B.subtilis and Sarcina lutea, and it has maximum antibacterial activity on S.aureus MR, but it has no antibacterial activity on P.aeruginosa. And aqueous extract from Girardinia suborbiculata C.J.Chen has no antibacterial activity on these several bacteria. 2. Antibacterial activity of Girardinia suborbiculata C.J.Chen and separated activity component(1) In MIC test on S.aureus MR, E.coli and B.subtili showed that the MIC of Girardinia suborbiculata C.J.Chen on three bacteria were between 10~1OOmg/ml.(2) In three extraction method include ultrasonic extraction, percolation extraction, reflux extraction, extract by reflux extraction has the maximum effect on anti-bacterial activity. (3) In different concentration of methanol and ethanol extracting test, only extract by 70% methanol and 70% ethanol have anti-bacterial activity, and 70% ethanol extract has the maximum anti-bacterial activity on S.aureus MR.(4) Different polar solvent extracting test show:anti-bacterial activity only existed in chloroform extract of ethanol extract from Girardinia suborbiculata C.J.Chen.(5) Separated anti-bacterial activity componentActivity component was separated by silica gel column chromatography from chloroform extract repeatedly, and prep-thin-layer chromatography (PTLC) was used for separating component with thin layer chromatography (TLC)-bioautography for separate instruction,at last. obtained 3 anti-bacterial active compound.(6) The structure analysis of compound:The structure of compoundⅢwas elucidated by infrared, electrospray ionization-mass spectrometry, nuclear magnetic resonance (NMR).
