Objective: To study the mechanism of a compound Chinese Medicine Yin-chen compound preparation on the Pharmacokinetics, Pharmacodynamics and Protecting liver. Baicalin of Yin-chen compound preparatio was determined by RP-HPLC assay; Pharmacokinetics of Yin-chen compound preparatio in mice and the Pharmacokinetic parameters were investigated; Protecting action and mechanism were studied of a Yin-chen compound preparation on the acute liver damage CCL4 and in vitro H2O2 liver cell damage; so as to provide scientific basis for its clinic application.Methods:(1) The RP-HPLC assay was invested to determine the content of baicalin in. Yin-chen compound preparation. The chromatogram condition was as follows: Hanbon-C18 column, the mobile phase was a mixture of methanol: water: phosphoric acid (47:53:0.2) with a flow rate of 1 mL/L and detection wavelength at 280nm. Temperature of column is 30℃.Baicalin was quantified with external standard method.(2) Mice of the Kunming were given various Yin-chen compound preparation (5.5g/kg, 11 g/kg, 16.5 g/kg) by i.g for sequence 4 days. A liver injury model was produced in these mice by injecting 0.5% of carbon tetrachloride-earthnut oil solution (0.1mL/10g) into the cervix on the 3rd days of the experiment respectively.. Effects of in-chen compound preparation on the liver histology, Alkaline Phosphatase( AKP), Alanine aminitransperase (ALT ), Aspartate aminotransferase(AST),Superoxide dismutase (SOD),glutanthione peroxidese(GSH-Px) and the concentrations of maleicdialdehyde (MDA) in the CCL4 intoxicated mice were kept under observation.(3) Pharmacokinetics of orally administered at a dose of 16.5 g/kg Yin-chen compound preparation were investigated in mice. Serum concentration of the effective component-baicalin in various times at 0.25, 0.5, 1.0, 2.0, 3.0, 4.0, 5.0, 6.0, 8.0, 12.0, 14.0, 16.0, 20.0, 24.0h were measured using HPLC and...
