Zoysia grass has quality of good palatability, soft, rich nutrition, is a good pasture, also a widely used warm-season turf grass. Because of its many excellent characteristics, such as strong adaptability, drought-resistance, extension ability, wear-tolerance, elasticity and et al., it is widely used in lawn, ecological restoration, water and soil conservation. However, it also has unfavorable aspects, relatively short green period and not too strong cold tolerance. Application of biotechnology in turf grass breeding can improve agronomic trait. The combination of traditional methods and modern breeding technique, especially modern biological technology, will shorten breeding cycle greatly. So using genetic engineering research on genetic improvement has attracted much attention. In this study we have established an efficient tissue culture regeneration system of Zoysia, through agrobacterium- mediated transformation BdDREB2 gene we lay the foundation for the cultivation of new varieties. The results were as follows:1. The optimum medium for callus induction using mature embryo of Zoysia as explant: MS+ 2,4-D(3 mg·L-1)+6-BA(0.1 mg·L-1); (Haiqing-1) MS+2,4-D(3 mg·L-1) +6-BA (0.2 mg·L-1); (Haiqing-2) MS+2,4-D(4 mg·L-1)+6-BA (0.2 mg·L-1); (Yishui Shandong and Xiuyan Liaoning) MS+2,4-D (2 mg·L-1)+6-BA(0.2 mg·L-1);2. The best medium for callus subculture: MS+2,4-D(2 mg·L-1)+6-BA(0.2 mg·L-1);3. The feasible differentiation medium: (Zoysia and Yishui Shandong) MS+KT(1 mg·L-1) +NAA(0.1 mg·L-1); (Haiqing-1) MS+KT(2 mg·L-1)+NAA(0.1 mg·L-1)+6-BA(1 mg·L-1); (Haiqing-2) MS+KT(1 mg·L-1)+NAA(0.1 mg·L-1)+6-BA(1 mg·L-1); (Xiuyan Liaoning) MS+ KT(1 mg·L-1)+ NAA (0.1 mg·L-1)+6-BA(1 mg·L-1);4. The rooting medium was 1/2MS+NAA (0.1 mg·L-1); Rooting ratio was 100%.5. PCR detection displayed that the exogenous BdDREB2 gene had integrated into the genome of Zoysia.
