Pleione bulbocodioides (Franch.) Rolfe is a famous orehids which can made Chinese traditional medicine Iphigenia indica with the function of Clearing heat, transforming phlegm,expelling Miasma and so on. Because of excess utilization of the resources, wildlife resources were stretcled to the limit. It had large number seeds which was a mass of undifferentiated embryogenic cell. Because the seeds were impervious to water and air and endosperm imperfectly developed, it hard to germinate under the natural condition.In this paper, with different culture medium, plants were regenerated at a high frequency using seeds as explants. Then induced Callus and adventitious bud using protocorms from seeds. A number of tube seedlings finally.The effects of concentrations of hormone, concentrations of activated carbon and sucrose on seed germination of Pleione bulbocodioides were investigated.The results showed that the optimal culture conditions are basal medium MS,1.0% sucrose and 0.5% activated carbon which had higher precentage germination. In the hormone experiment,1.5mg/lIAA had the best result whose gemination rate was 6.5%.Different concentration of IAA had better precentage germination which induced more and larger protocorms whose average Diameter lmm≦d≦1.5mm.The medium with NAA is better suited to root. Morphological observation showed that the process that Embryo Cells broke through the testa to protocorms, then complete plant had regularity.The callus induction of Pleione bulbocodioides (Franch.) Rolfe was studied with protocorms from seeds as explants. The effects of concentrations of hormone, types of auxin and cytokinin, concentrations of activated carbon and sucrose on callus induction of Pleione bulbocodioides were investigated. Callus induction rate, browning rate and the degree of callus-forming were considered as indicators. The results showed that the medium which induced callus was optimal with 1/2MS+1.0 mg/l2,4-D+1.0mg/l 6-BA+2% sucrose+ 0.1% activated carbon+0.55% agar.
