Ning Qianhu(Peucedanum praeruptorum Dunn),the umbelliferae peucedanum perennial herbaceous plants,of which the dry storage root is commonly used in traditional Chinese medicine with the function of dealing with anemopyretic cold,cough with abundance of phlegm.And often used for the treatment of phlegm heat cough,asthma and bronchitis airway disease.The main propagate methods of P.praeruptorum Dunn is seed germination.Seed germination is of great significance in the process of plant growth and development,its germination quality directly affects the growth and development of plants and the ability to adapt to the environment.In recent years,due to the blind exploitation of wild P.praeruptorum Dunn resources,resulting in the reduction of wild resources,resulting in the natural resources of wild P.praeruptorum Dunn endangered.On the other hand,a variety of problems such as germplasm degradation,pesticide residues and seed borne diseases were found in the wild P.praeruptorum Dunn cultivars.In addition,the species and quantity of medicinal active components of artificial cultivated P.praeruptorum Dunn are often different because of different ecological conditions,such as habitat and climate,these conditions have brought many difficulties to the quality and quality control of commercial medicinal materials.In this study,P.praeruptorum Dunn seeds,leaves,stems and tuberous roots were used as experimental materials to study the characteristics of seed germination and tissue culture regeneration system.The main results were as follows:1.The effects of exogenous hormones ABA and its inhibitors FL and NDGA,GAS and its inhibitors PA on seed germination of P.praeruptorum Dunn were studied.Our results showed ABA is the main reason for the dormancy of P.praeruptorum Dunn seeds,while GAS is necessary for germination.The application of ABA and its inhibitors,FL and NDGA,can significantly improve seed germination rate of P.praeruptorum Dunn and its seedling tidy.This result has an important guiding role in the actual production.2.The effects of exogenous hormones ABA and GA3 on the physiological and biochemical indexes of P.praeruptorum Dunn seedlings were studied.The results showed that seed soaking with ABA could efficiently improve the stress resistance index of seedlings,but the weaken seedlings were serious,which was not conducive to the accumulation of organic matters,while seed soaking with GA3,it makes seddlings healthy and strong,the accumulation of organic matters and plant height were significantly increased,and it is important to improve the quality and yield of P.praeruptorum Dunn.3.Study on the establishment of in vitro sterility culture system of P.praeruptorum Dunn,leaves as explant is the optimum selection in three types of explants,like leaves,stems and tuberous roots of P.praeruptorum Dunn,and secondary choice followed by stems and tuberous roots.4.NAA 0.5 mg/L + TDZ 0.05 mg/L is most optimal for leaf callus induction of P.praeruptorum Dunn,the highest induction rate was 95.56%.Dark or low light intensity was beneficial to the formation of leaf callus,and the callus appeared for the first time after in vitro cultured 8 days.Leaves abaxial surface side down to contact medium was better than adaxial side down culture for callus formation.NAA 0.5 mg/L + 6-BA 2.0 mg/L is most optimal for stem callus induction of P.praeruptorum Dunn,the highest induction rate up to 80.00%.The stems horizontally placed on medium is conducive to the formation of callus,vertical placement(morphology bottom insert to the medium)was beneficial to the differentiation of adventitious buds on the top of the morphology of stem,and the callus was formed at the boundary between the morphology bottom and the culture medium,but not differention.NAA 0.5 mg/L + 6-BA 2.0 mg/L or 2,4-D 2.0 mg/L + KT 0.5 mg/L is most optimal for tuberous root callus induction of P.praeruptorum Dunn,the highest induction rate was 78.89%.5.MS + 6-BA 1 mg/L + NAA 0.5 mg/L had the optimum effect on P.praeruptorum Dunn callus proliferation and differentiation,and the differentiation rate was as high as 66.67%,the proliferation coefficient was about 4.63,the callus growth was better and the volume was larger.6.The optimum seedling rooting medium for P.praeruptorum Dunn was 1/2MS + NAA 0.5 mg/L + 6-BA 0.2 mg/L,the highest rooting rate reached 41.1%,the average root number is 7.2,the growth of root is thick and strong,root length approximately 8 ~ 11 cm;vermiculite: mixed matrix humus soil =1:1 matrix is the optimum cultivation of P.praeruptorum Dunn tissue culture seedlings,and the transplanting survival rate as high as 95.3%.
