This paper taking four sainfoin with large cultivated area and wide application in production as the experimental materials. This stidy use the orthogonal experiment combined with visual analysis method to study the influence of different types of explant, various hormone and hormone concentrations on callus induction and plant regeneration of the germplasms. The results show that:1. In 5 kinds of explants (root, hypocotyls, cotyledon, stem and leaf), hypocotyl showed high stability in both callus induction and somatic embryo formation ability, which proved to be the best explant.2. The main factor that influence the formation of callus induction in 4 sainfoin materials is 2,4-D. The best hormone ratio of callus induction is 6-BA 1.0mg/L+NAA Omg/L+2,4-D 2.0mg/L+KT 1.0mg/L for Onobrychis viciaefolia Scop.cv.Mengnong, 6-BA 0.1 mg/L+NAA 0.01mg/L+2,4-D 1.0mg/L+KT 0.5mg/L for Onobrychis viciaefolia,6-BA 0.5mg/L+NAA 0mg/L+2,4-D 1.0mg/L+KT 0.5mg/L for Onobrychis viciaefolia Scop.cv.Reymont,6-BA0.5mg/L+NAA 0mg/L+2,4-D 4.0mg/L +KT 0.5mg/L for Onobrychis viciaefolia Scop.cv.Eski.3. The main factor that influence the formation of somatic embryos in 4 sainfoin materials is NAA. The best hormone ratio of callus induction is 6-BA 2.0mg/L+NAA 0.1mg/L+KT 0mg/L+CH 1.0g/L for Onobrychis viciaefolia Scop.cv.Mengnong,6-BA 2.0mg/L+NAA 1.0mg/L+KT 0mg/L+CH 2.0g/L for Onobrychis viciaefolia, 6-BA1.0mg/L+NAA 0.1mg/L+KT0.5mg/L+CH 0g/L for Onobrychis viciaefolia Scop.cv.Reymont,6-BA 1.0mg/L+NAA 0.1mg/L+KT 2.0mg/L+ CH 2.0g/L for Onobrychis viciaefolia Scop.cv.Eski.4. The comprehensive analysis of this study showed that the best hormone ratio for callus induction is 6-BA(0.1~1.0mg/L)+NAA(0~0.01mg/L)+2,4-D (1.0~4.0mg/L)+ KT(0.5~1.0mg/L). While the best hormone ratio for somatic embryo induction is MS+ 6-BA(1.0~2.0mg/L)+NAA(0.01~1.0mg/L)+KT(0~2.0mg/L)+CH(0~2.0 g/L).
