Effervescent tablets of Oxytetracycline Hydrochloride were preparated in this study. It's quality control, the minimum inhibitory concentration to bacteria, the protective effect to the chickens infected bacteria in artificially , and the LD50 to the chicken were studied.1. Prescription and production processThe formulation of Oxytetracycline Hydrochloride effervescent tablet was stablished through changing the auxiliary selection and the acid-base ratio. The composition of Oxytetracycline Hydrochloride, sodium carbonate, citric acid, hydroxypropyl methyl cellulose (HPMC), lactose, polyethylene and glycol 6000 in formulation is respectively 15%, 31.5%, 22.8%, 3.5%, 24.6%, 2.8%.2. Quality ControlQuality test of Oxytetracycline Hydrochloride effervescent tablets: the medicament could completely was disintegrate with in 5 min,when pH value between 4.0 and 5.5, the average volume of foam was larger than 12.63 mL, and the average disintegration time was 4.52 min, hardness was 10.58 Kg/cm2, all these consistent with the standard of China pharmacopeia. The average recovery rate was 95.07% using UV spectrophotometry to determine the medicament content, and the results indicated that the test method was accurate and reliable. Stability test showed the Oxytetracycline Hydrochloride effervescent tablet was sensitive to the humidity and light, but insensitive to the temperature.3. Pharmacodynamic Study of Effervescent tablets of Oxytetracycline HydrochlorideThe study of effect on Oxytetracycline Hydrochloride effervescent tablet was carried out by using in vitro antibacterial experimentation and protection test to artificially infected chickens.Antibacterial test showed theMIC (Minimal inhibitory concentration ) of Oxytetracycline Hydrochloride effervescent was respectively 4μg / mL, 16μg/mL, 2μg/mL, 2μg/mL and 8μg/ mL to E. coli, Streptococcus agalactiae, Staphylococcus aureus, Salmonella, Pasteurella.This drug have therapeutic effects on artificially infected chicken with E. coli, S. aureus and salmonellas. Mortality of the treatment group was significantly lower than the infected control group (P<0.05). The high dose efficient is 70%, medium dose efficient is 66.7%, low dose efficient is 57.8% to chicken infected with E. coli. The high dose efficient is 65%, medium dose efficient is 63.2%, low dose efficient is 45% to chicken with Staphylococcus aureus. The high dose efficient is 57.8%, medium dose efficient is 52.6%, low dose efficient is 55% to chicken with salmonella. Conclusions:It had better therapy effect than the Oxytetracycline Hydrochloride tablets.4. Determination of LD50 doseThe LD50 of Oxytetracycline Hydrochloride effervescent tablet to chicken was 1440mg/kg calculated according to Karber method, and its 95% confidence limits were between 1247 mg/kg and 1663 mg/kg, and the average of its 95% confidence was1440±208 mg/kg. The results showed concluded that this medicine is a safe antibacterial agent with low toxicity.
