In recent years,under the social demand of healthy breeding and ensuring food safety,the problems of antibiotic residues,drug resistance,toxic and side effects have attracted people’s attention.Countries around the world have begun to ban the addition of antibiotics to feed.With the proposal of "feed ban" and the development of animal husbandry in China,green feed additives in feed will have great development space.Probiotics is a new type of feed additive with no toxicity,drug residues or drug resistance,and is expected to become one of the effective alternatives to antibiotics.At present,probiotics commonly used in the market include lactic acid bacteria,bacillus,photosynthetic bacteria and fungi,and bacillus has remarkable development advantages and application prospect due to its excellent characteristics of strong stress resistance,environmental adaptability,high temperature and high pressure resistance,convenient storage and the like,as well as regulation and improvement of intestinal flora,improvement of animal immunity,improvement of animal production performance and improvement of breeding economic benefits.At present,bacillus preparations also have the problems of hemolytic activity,toxin gene,cytotoxicity,drug resistance and the like,and the development of the bacillus preparation which is convenient to use becomes an urgent problem to be solved in production.This test from the traditional fermented food and sensitivity to antibiotics was isolated from soil samples,good tolerance,good features good bacillus strains,through experimental prove strains of salmonella infection in mice live action,will get the good strains of filtering processing preparation of effervescent tablets and optimize its preparation technology,the effervescent tablets in late egg laying hens in drinking water,To investigate the effects of Bacillus on performance,egg quality,serum antioxidant capacity and immune function of laying hens in late laying period.The main results are as follows:(1)A total of 96 strains of Bacillus were isolated from a large number of fermented food and soil samples.Nine strains of Gram-positive rod-shaped bacteria were screened by Gram staining,acid and alkali tolerance,bile salt tolerance and trypsin tolerance tests,which were resistant to strong acid(pH=2),strong alkali(pH=12),bile salt tolerance(0.2%)and trypsin tolerance(0.1%).(2)Four dominant strains were screened out by bacteriostatic test and drug sensitivity test,which could inhibit common pathogenic bacteria and be generally sensitive to antibiotics.(3)According to the molecular biological identification,among the four strains,K52 and K26 are Lysinibacillus macroides,K8-2 is Bacillus halotolerans,and K7 is Bacillus licheniformis.The safety of these four strains is verified by mouse safety test.(4)Through the anti-salmonella infection test in mice,the anti-salmonella ability of the four strains was compared,and bacillus lysine K52 with strong probiotic performance was selected for the subsequent preparation of Bacillus effervescent tablets.(5)The acid source,acid-base ratio,lubricant and filler of effervescent tablets were determined by single factor test.L9(34)orthogonal experiment was designed on the basis of single factor experiment,the acid-base ratio,PEG 6000 addition amount and mannitol addition amount were selected.The optimal formula was determined by calculating comprehensive score according to foaming amount,disintegrating time and pH value,which indicated that citric acid was the acid source and mannitol was the filling agent.The acid-base ratio was 1:1.5,PEG 6000 dosage 5%,mannitol dosage 20%.(6)The probiotic effect of Bacillus lysine K52 was comprehensively evaluated by adding the prepared Bacillus effervescent tablets into the drinking water of laying hens at the late stage of laying.The effects of the tablets on the behavior,performance,egg quality,serum antioxidant level and immune function of laying hens were investigated.The results show that:a Bacillus a significantly increased the laying rate of laying hens in late laying period(P<0.05),there were no significant differences in average egg weight,feed to egg ratio and average daily feed intake(P>0.05);b Bacillus b significantly increased eggshell weight and strength,egg weight,protein height and Hastelloy unit(P<0.05),egg shape index,eggshell thickness and yolk relative weight had no significant effects(P>0.05);c Bacillus c significantly increased the levels of SOD and T-AOC in serum of laying hens during late laying period(P<0.05),decreased MDA content;d Bacillus improved the immune effect of the trivalent inactivated vaccine of recombinant avian influenza,and improved the humoral immunity and cellular immunity of laying hens in the late laying period.At the same time,it was found that the effect of feeding Bacillus at the same time of immunization was better than that of vaccination after feeding Bacillus first.The test results proved that Lysinibacillus macroides K52 had the potential to develop multifunctional microecologics,which was expected to be used in the subsequent development and application.
