In the process of maintenance, the practice of regeneration by multiplication of a sample of each accession in small field plots as well as seed aging, leading to a serious threat to the purpose of genetic integrity. Therefore, germplasm conservation has attracted much attention.In this study, the genetic integrity of the Bromus stored in Innermongolia was investigated by ISSR. The analysis of seed germination identifies genes related to conserving safly. The results show as following:The genetic integrity of 14 accessions of Bromus was analyzed by 20 ISSR primers. Accessions each were represented by two sub-populations regenerated by different conserving methods. The results of ISSR data analysis is that: on the 20 ISSR loci, a total of 150 alleles were detected in these 14sub-populations. At each locus, 4 to 9 alleles were detected. It included 4 to 14 alleles were detected; also it included 4 to 26 alleles were detected.Genetic similarity analysis showed that: the genetic similarity coefficients of each two sub-populations regenerated by different conserving methods differ from 0.78to 0.93, except Bromus inermis Leyss(3) . While Genetic similarity of the Bromus remotiflorum (Stend.) Ohwi(1) and Bromus inermis Leyss(3) are both above0.93.Banking conserving lost less gene than the maintenance by the outside. so it should be different between banking conserving and gmplasm nursery conservation. The genetic integrity could be changed when the water content of Bromus cilatus was above 8.5% and Bromus sadi was about 7.67%.so lowing the water content of materials could conserve seeds more safely. However different materials have different right water content.
