Using the progeny of sorghum 314A×brown shell sudangrass in the F2: 3 progeny of genetic mapping population build Recombinant inbred populations F7:8.,in the group of the leaf length, leaf width, ear length, tiller number and plant fresh weighs 5 traits by two methods, Based on characters of NILs isolation and based on molecular markers of NILs separation, separated and obtained the 5 traits of near-isogenic lines two traits are Effect of interaction sites NILs, in addition separated and obtained optimized Sorghum SSR system, the main results are as follows:1 A number of factors were studied to set up the optimizing SSR reaction system. The optimized technology was: Each 20μl amplification reaction solution comprised DNA(60ng )1.0μl,Mg2+(25mM)2.0μl,Tap(5U/u1)0.25μl,dNTPs(2.5mM)2μl,Primer(10uM)1μl, 10×Buffer 2.5μl,ddH2O 10.75μl。This system had well reproducibility and stability proved by amplification practice.2 In this study, sorghum 314A×brown shell sudangrass in the progeny of genetic mapping population build recombinant inbred populations were as material, Through methods those based on phenotypic traits and molecular markers reciprocal combination to separate NILs, removed NILs those leaf length, leaf width, ear length, tiller number and plant fresh weighs from Genealogies of RIL-82, RIL-97, RIL-151, RIL-117, RIL-154 and RIL-132.3 NILs those nteraction sites (phi024-phi053) of tiller and leaf width and Interaction sites (phi024-phi053) of leaf length and fresh weight per plant were Separated from Genealogies of RIL-68 RIL and RIL-141.4 This reserch not only gets the near-isogenic lines in the seperation traits also gets nine high-producing Recombinant inbred populations offsprings.Although these offsprings exist some similarities in the main agronomic characters, there are significant differences.in the resistances.At present ,we have screened four kinds of new Sorghum germplasm resouces with excellent characters by the appraisal of agronomic characters and resistances .Meanwhile , two kinds of the germplasm resouces are conducting the regional tests. Therefore, using two kinds of methods for restructuring inbred lines of NILS separation not only can shorten the gap between the reseach of gene location and application of breeding also can reduce the breeding period and enhance the benefit of selection.5 Using NILs those Obtained from Separation to locate QTL trait loci of propertie and to Separate QTL trait loci and to analyze genetic effect of Interaction sites(epistasis).
