This paper introduces an efficient method for isolation of mitochondrial DNA (mtDNA) from mycelium of Volvariella volvacea. The mitochondria isolation used different centrifu gation from the cell. To isolate mtDNA , mitochondia were lysed with SDS and proteinase K after digesting by DNaseI. Mitochondrial DNA were separated from protein and RNA with TE-saturated pheol/chloroform. Evaluation of mtDNA by agrose gel electrophoresis showed that the quality of the final mtDNA met the experiment's demand of genetic studies.From the result of Volvariella volvacea mitochondrial genome,we got 41 Scaffolds,152 Contigs,the sequcence bases of them is about 100kb,and in which the Scaffolds and Contigs that more than 1kb is respectively 24 and 57,the length of them is respectively 46678bp and 38193bp.Sequence fragments were used for codon, base composition, endonuclease digestion and repeat sequence analysis,the resulted showed that: genes had a preference for the codon with A or T as its third base in V.volvacea mtDNA; the AT base content was higher than GC base content; there were 14 one cleavage frequcney site in the 2 scafolds sequence fragments; there was one type of 6 base repeat unit, four types of 8 base repeat unit, one type of 9 base repeat unit, two types of 11 base repeat unit and two types of 19 base repeat unit, and the type of 20,21,23,29,42,84 base repeat unit is respectively one.14 protein coding genes, 2 rRNA genes and 21 tRNA genes were found by blast.Compared with eight kinds of Basidiomycetes mitochondrial genomes, V.volvacea has the same genomes composition, the gene order has some some regularity.For the V.volvacea classification and identification, the protein sequences of 27 mtDNA were analysis by ML and BI miethod .The result shown that they have the simliar taxonomic status to the traditional method.
