According to the distribution status of the germplasm resources of Eremochloa ophiuroides from different provinces:Hainan, Guangdong, Fujian, Zhejiang, Jiangsu, Anhui, Henan, Hubei, Jiangxi, Hunan, Chongqing, Sichuan, Yunnan, Guizhou and Guangxi province in China, investigation and collection was made and materials were analyzed in two major levels:morphology level and DNA level, to test the genetic diversity of 60 samples of Eremochloa ophiuroides germplasm resources that gathered from above thirteen provinces. Integrated analysis of morphology level was made in two parts:space enlargement of clones and turf quality, these analyses provided basis for study, collection, new cultivates breeding and rational utilization of the germplasm resources of Eremochloa ophiuroides.(1).Research results of Eremochloa ophiuroides morphological markers:Among the 11 morphological markers, the maximum range variation was in the leaf length of vertical branch, which was about 22.15%; the minimum range variation was existed the diameter of stolon, which was only 7.82%.Of 12 characteristics of stolon horizontal enlargement, correlation analysis showed a highly significant level (P<0.01) of stolon stem length, internodes length, stolon leaf numbers, tiller numbers, second-level internodes numbers, first-level nodes length. The stem internodes length, first-level branch internodes numbers and second-level internodes length showed a significant level. The variation range of each marker was 0.51%-3.28%, which means the rich diversity among different asexual propagation characterization markers.(2).Assessment of sixty germplasm of Eremochloa ophiuroides using the Comprehensive Assessment System of Turf Quality proposed by Liu Jianxiu. The results showed that among different varieties of turf grasses, NO.54 germplasm scores the highest, while NO.48 scores the lowest, the leaf morphology index value ranges in the middle level in the rest of germplasm, the evaluation score proclaimed that Eremochloa ophiuroides is suitable to different kinds of turf.(3).Genetic diversity of sixty germplasm of Eremochloa ophiuroides using ISSR molecular marker analysis. Thirteen highly polymorphism and resolution pairs of ISSR primers were used to the amplification of genome DNA of materials,104 pieces of readable bands were obtained, including 96 polymorphic bands; the polymorphic sites percentage was 92.31%, which means the sweeping variation in DNA level of materials. Tested by NTSYSpc2.10e software, the average valid number of alleles locus, the average diversity and the average Shannon information index was 1.468±0.129,0.277±0.064,0.425±0.085, separately. The genetic similarity coefficient was between 0.5691-0.9106, and the average was 0.7396; The sixty germplasm were divided into two groups using UPGMA method.
