Mass Production Of Protocorm-Like Bodies With Bioreactor And Establishment Of Regeneration System In Cymbidium Grandiflorium

Cymbidium grandiflorium is one of the advanced flowers which have higher ornamental values. Process of scale production was greatly influenced because of the low rate of germination and low speed of division propagation. In this study, stem apex of C. grandiflorium was used as explants to investigate the factors of affecting the induction and proliferation of protocorm like body (PLB), proliferation of shoots and rooting of C. grandiflorium in vitro. In addition, the effects of several the factors on PLB proliferation and growth during air lift bioreactor culture were examined, the system of tissue culture and rapid propagation for C. grandiflorium was also discussed, which provides the theoretical basis for large scale production of C. grandiflorium.The results were as follows:During the culture of PLB proliferation for C. grandiflorium in vitro, MS medium supplemented with 0.2 mg·L-1 NAA and 1.5 mg.L-1 BA was the most favorable for PLB proliferation, inoculation of 20 g was better than 10 g and 30 g in 5 L bioreactor for PLB proliferation,30 g-L"'of sucrose was helpful for PLB proliferation, and the optimal concentration of active carbon(AC) for PLB growth was 0.25 g·L-1 and higher concentration of AC restricted the growth of PLB. In addition, adventitious shoots were induced effectively in MS medium supplemented with AC 1.0 g·L-1, BA 1.5 mg·L-1, coconut 100 g·L-1 and NAA1.0 mg·L-1, adventitious shoots grew well in MS medium supplemented with BA1.0 mg·L-1 and NAA 2.0 mg·L-1, and the optimum rooting medium of adventitious shoots was MS medium containing IBA 0.5 mg·L-1,and 95.3% of rooting was obtained.
