Taihang black goat is the local goat, the clothing hair is mixed types, the coat color is black and bright with slender purple cashmere fibre and the influence of artificial selection is lower with significant breed characteristics. In this experiment, the CDs region of Fibroblast Growth Factor5 (FGF5) gene was cloned from total RNA by RT-PCR, the sequence characterization and expression of follicle growth in different development period of FGF5 gene were performed with it. The result showed:1. The CDs region of FGF5 in Taihang black goat consisted of 813 nucleotides encoding 270 amino acids with a FGF domain from 87-221 amino acids. The nucleotides sequence homology of FGF5 CDs of Taihang black goat was found to be 98%,92%,90%,90% and 86% compared with that of bovine (NM001078011),horse (XM001492556),human (NM004464.3),orangutan (XM526572.2) and mouse (NM010203.4), while the amino acid sequence homology was 98%,91%,91%,91% and 91% respectively. The amino acids coded by the gene FGF5 are highly conserved in Taihang black goat;2. In the all amino acids consisting of FGF5, the relative content of Ser,Gly,Lys,Leu,Ala and Pro was higher than the others. Its relative molecular weight was 29540.7Da, the isoelectric point was 10.67; the total number of positively charged residues (Asp + Glu) was 15, the total number of negative charge residues (Arg + Lys) was 43; the molecular formula was C1312H2081N391O376S6, The total atomicity was 4166, the half-life was about 30 h. The instability index is computed to be 57.50 and the protein is unstable, the aliphatic index is 63.26. Its maximum of hydrophilic was 3.067, the biggest peak of hydrophobicity was -2.789; It was a secretory protein, contained 2 transmembrane domain. The FGF5 protein was containing 25 Ser, 2 Thr and 3 Tyr, which probably be protein kinase phosphorylation sites. The prediction of Signal peptide analysis results showed: the 1~21 amino acids in N-terminal was a classical signal peptide sequence;3. In the detection of four months skin tissue, the relative expression of July, August, September and October in FGF5 were (0.019906±0.003a), (0.015713±0.001a), (0.028374 ±0.001b) and (0.019466±0.002a). The result speculated that FGF5 gene may play a role as a continuous expression pattern model in Taihang black goat. The statistic analysis showed that: it was highly expressed in September, significantly higher than July, August and October (P<0.05), but there is no significant differences between these three months (P>0.05); The result of real time quantitative indicated that the follicle development of Taihang black goat was in anagen in July and August, the followed was the transition of anagen and catagen in September, and enter into catagen in October.
