Dairy Cow Mastitis is an important disease that greatly restricts the development of dairy industry, and has become the most expensive disease by its high incidence, low cure rate and complex pathogenesis. In order to reduce mastitis on the dairy industry losses, to ensure the health and sustainable development of dairy industry, so in this study we detect serum inflammatory factors, and through biochemical analysis, to screen the factors that have close relationship to Mastitis in Dairy Cattle, then provide the basis for the in-depth research on physiological metabolism of cow mastitis. And trying to find a more convenient, economical, rapid and reliable detection index, which are the foundation of the model that using serum biochemical components change to quickly diagnosis the cow suffering mastitis or not.In addition, this study also hopes through the analysis of cow genetic structure to find out the susceptibility and resistance-related genetic markers for assisted selection, and thus to reduce losses and promote animal husbandry development, then protection of human health. Of course, this experiment also utilization of a variety of bioinformatics software to clarify the influence on single nucleotide polymorphism, arising some interpretation and explanation on SNP to some extent.1.The detection of serum inflammation-related factor Use biochemical analyzer, detect some serum indexs (include Hs-CRP,ALB,AAG,PA,ALT,AST,TBIL,GLU )that related to infection. It was found that the serum level of ALB and ALT are significant difference between the normal group and infected group (P<0.05), especially the ALB, which is greatly lower in affected group (p<0.01). Other serum indexs, such as Hs-CRP, ALT, GLU, TBIL have no significant difference. It is evident that ALB and ALT can be the cow mastitis serum indicator.2. The research on CRP gene polymorphism Detect the polymorphism in C-reactive protein (CRP) and analyze the relationship between CRP gene and mastitis (including mastitis resistance, susceptible, milk quantity and quality). According to the sequence of bovine CRP gene, we design four pairs of primers and use PCR-SSCP to detect single nucleotide polymorphism of CRP gene in Chinese Holstein, and analyze the relationship through the software SPSS and Excel. There were polymorphic locus occurring in P1,P2 and P3,but the mutant locus found in P1 and P2 are the same. P3 have two genotppe (TT, GG), and P1 (P2) have three genotypes (AA, AB, BB), but no polymorphic locus occurring in P4. The relationship analysis indicate that the genotype and the mastitis have no significant difference, and also do little to the milk quality and quantity, but all of the mastitis dairy are AA or TT genotype.3. The bioinformatics analysis of CRP gene SNP Analyze the influence of single nucleotide change by network software. The results showed: there is SNP (G→T mutation) in CRP exon 2, and the mutation make amino acids Q→H change. Through bioinformatics analysis we found that this change leads to a new combination point of transcription, which called vaccinia-term-s. But the mutation did not cause other changes. This result can provide, to some extent, an explanation on the relationship between CRP gene mutation and the function change.
