Drought stress is the main abiotic stress that harmful to crops. in order to explore wheat drought resistance mechanism the test use physiological and biochemical to research wheat metabolism of drought stress. Experiment with YanD27, Shanyou 225, Xinmai 18, Xiaoyan 22 and 10-31 as tested materials investigate the wheat germination rate, germination energy, coleoptile length, and relative water content(RWC). By 18%, 35% of the two concentrations of PEG-6000 treatment for wheat seedlings 24h, SDS-PAGE protein separation, analysis and comparise in protein quantity and quality. HPLC-MS/MS mass spectrometry revealed that the protein expression difference on quantity and quality in drought environment. The primary results are:1. Simulated drought stress affecte germination, germination, coleoptile length and relative water content of different varieties. Drought-resistance varieties' germination rate and germination, coleoptile length and relative water content has little effect when drought stress; non-drought-resistant varieties' germination rate and germination , coleoptile length and relative water content are impacted greatly.2.SDS-PAGE showed that at different concentrations of PEG-6000 the protein abundance increase in drought-resistance varieties Yan D27, Shanyou 225; the protein abundance increase in non-drought-resistant varieties Xiaoyan 22, the protein abundance of Xinmai 18 resistance;10-31 protein abundance reduce suggested with weak drought.3.HPLC-MS/MS identificate different protein in the SDS-PAGE of Xinmai 18 and found four types of proteins, enzymes and proteins and photosynthesis, and respiration-related enzymes and proteins, regulatory proteins, and a species of unknown proteins. Regulatory protein 14-3-3 proteins related to drought stress.Differences in proteomics perfect the wheat drought molecular's resistance mechanism.proteomics,morphology and physiological indexes provide the theory basis for identification of drought-resistant wheat.
