Veterinary drugs are applied in animal husbandry to promote animal growth,prevent infection and treat diseases.However,the improper and illegal use of prohibited or permitted drugs may result in the presence of drug residues in the animal tissues,leading to serious potential hazards to consumer.Currently,the high-throughput screening and confirmation technology used to monitor multi-class drug residues in animal-derived foods still faces many challenges,such as the low concentration of substance,the complexity of the biological matrix and because of the interferences.Therefore,the establishment of a fast,effective,economical,high-throughput and wide-coverage method of screening drug residues in animal food is imperative.In this paper,several ultra high-performance liquid chromatography tandem mass spectrometry method was developed for the high throughput screening and quantitative determination of multi classes drugs in animal derived food.The target drugs include pesticides,veterinary drugs and other drugs.The detailed chemical classes include macrolides,sulfonamides,tetracyclines,?-lactams,?-agonists,aminoglycosides,antiviral drugs,glycosides,phenothiazine,protein anabolic hormones,non-steroidal anti-inflammatory drugs(NSAIDs),quinolones,antifungal drugs,corticosteroids,imidazoles,piperidines,piperazidines,insecticides,amides,alkaloids and others.The specific works are summaried as follows:The multi-residue determination of 210 drugs in pork by ultra-high-performance liquid chromatography-tandem mass spectrometry was developed.A rapid and simple preparation method was applied to process pork sample,including solvent extraction with an acetonitrile/water mixture(80/20,v/v),defatting and clean-up procedures.The recoveries ranged from 52% to 130% with relative standard deviations(RSDs)< 20% for three spiked concentrations of 10,50 and 250 ?g/kg.More than 90% of the analytes achieved low limits of quantification(LOQs)< 10 ?g/kg.The decision limit(CC?),detection capability(CC?)values were in the range of 2-502 ?g/kg and 4-505 ?g/kg,respectively.The results show that the method has good accuracy and precision.The multi-residue determination of 65 drugs in eggs by ultra high-performance liquid chromatography-tandem mass spectrometry was eatablished.The QuECh ERS extraction method and a new nano-adsorbent material were integrated into the pretreatment process.The recoveries ranged from 50% to 130% with RSDs < 20% for three spiked concentrations of 1,10 and 100 ?g/kg.The LOQs ranged from 0.006 to 0.95 ?g/kg.The CC? and CC? values were within the ranges of 2-87 ?g/kg and 3-173 ?g/kg,respectively.The new nano materials provide a new method for the pretreatment of samples,and provide a new way for the screening of other animal derived food residues.The multi-residue determination of 95 drugs in eggs by ultra high-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UHPLC-QtoF)was developed.The QuEChERS extraction method and a new nano-adsorbent material were integrated into the pretreatment process.The recoveries ranged from 50% to 130% with RSDs < 20% for three spiked concentrations of 5,10 and 100 ?g/kg.The LOQs ranged from 0.12-14 ?g/kg.The CC? and CC? values were within the ranges 17-79 ?g/kg and 29-149 ?g/kg,respectively.The method provides a high resolution mass spectrometry detection method for the screening of multi-classes drugs in poultry products.A novel workflow strategy was developed to simultaneously screen multi-class drugs in animal derived food by combining UHPLC-QToF-MS and UHPLC-MS/MS.The proposed approach comprises three stages:(1)automated screening using UHPLC-ToF-MS in ToF-MS-IDA-MS/MS mode;(2)identification using UHPLC-ToF-MS with criteria including retention time,mass accuracy,isotope pattern,and MS/MS library search results;and(3)confirmation and quantification using UHPLC-MS/MS in multiple reaction monitoring(MRM)scheduled acquisition mode.Our goal was to develop a representative thought process enabling other researchers to establish a suitable method for their own needs in accordance with the proposed workflow.
