Objective: In order to investigate the relationship among antioxidant complex (vitamin C, vitamin E and polyphenol),free radical metabolism and physiological signs, to discuss free radical metabolism influence canine immunity, digestibility as well as the health of canine. The level of antioxidant complex was regulated. So we can enhance the health condition and welfare of canine as domestic animal through the nutrition regulation. Methods: The experiment selected twelve 60±15 days old SD beagle puppies, which were randomly divided into two groups: one control group and one experiment group. There were 5 female and 1 male in each group. All puppies were living by cage rearing. The control group was fed a basal diet with high protein and high fat. At the basal diet lever, the experiment group was supplied with antioxidant complex (vitamin C, vitamin E and polyphenol) mixed in basal diet. Each group's growth and hair condition were observed during the experiment stages and conducted the blood routine examination and biochemical analysis (ALT, AST) on day 1, day 29 and day 57 during this experiment. The concentration of SOD, MDA, NO in puppies'serum was also identified on day1, day29 and day 57. Meanwhile, the digestibility of each group from day 50 to day 54 was also test. Results: The dietary added antioxidant complex (vitamin C, vitamin E, polyphenol) affected the metabolism of free radical in canine. Activity of SOD was enhanced. The level of MDA and NO were reduced. On day 57, activity of SOD in the experiment group was higher than the control group by 7.18% and significant difference (P<0.05) was observed.Antioxidant complex also improve the immunity of puppies in the experiment group. The level of WBC, lymphocyte and neutrophilic granulocyte were decreased into normal physiological range on day 29. And the percentage of lymphocyte and granulocyte also turned normal at the same time. Significant difference (P<0.05) of immunity level was observed on day 57 between the control and experiment group.Antioxidant complex improved the digestibility of crude protein by 2.42%. It also maintained the level of ALT and AST within normal range, made daily gain of the beagle puppies increased; made hair condition obviously improved, particularly in getting rid of scurf. Conclusion: Antioxidant complex (vitamin C, vitamin E and polyphenol) can increase the canine immunity, repair the oxidative damages and also eliminated the free radical. It is playing an important role of balance the oxidized and the anti-oxidized effect. It can improve the health condition of canine including digestibility safety, daily weight gain and hair condition as well as maintain normal physiological signs. It also showed that certain ability of the anti-oxidized.
