| The porcine oocytes and embryos in vitro culture were the basic link of the in vitro production, and were the prerequisite in vitro fertilization,transfer gene and nuclear. So, the factors on in vitro maturation and parthenogenetic embryos of porcine oocytes were observed, In order to find the best in vitro culture system. The results as follows:1 Addition of 0.1% PVA and 10% FBS in maturation medium had no significant effect on mature rate,cleavage rate of 36h and day 6 morula rate(P> 0.05);Addition of different concentration of EGF in maturation medium had no significant effect on mature rate(P> 0.05),but addition of EGF increased the cleavage rate and morula rate of parthenogenesis embryos, and addition of 30ng/mL EGF is best for parthenogenetic embryos of development. addition of ITS to the maturation media could increased the morula rate of parthenogenesis embryos, but restraind the mature rate and cleavage rate.2 Different culture media had different effection on the porcine parthenogenesis embryos of development after activation. The NCSU-23 supplemented 0.4% BSA was a better culture media for the oocytes development after activated in the early stage, mTCM-199 supplemented 0.1% PVA facilitates parthenogenesis embryos breakthrough 4-cell block and reach morula. Sequential culture system of NCSU-23 supplemented 0.4% BSA and mTCM-199 supplemented 0.1% PVA were suitable for parthenogenesis embryos in vitro culture.3 The concentration of CO2 had no significant effect on mature rate(P> 0.05),but had a significant effect on cleavage rate. The cleavage rate and morula at 4% CO2 were significant higher (P<0.05)than that of the others; In the hypoxic conditions, addition of FSH with LH and LHRH-A3 were significant lower(P<0.05) than PMSG with HCG of the mature rate, but had a significant (P<0.05)effect on the cleavage and morula rate, and addition of the FSH with LH had a significant (P<0.05)effect relative LHRH-A3. There was no significant effects on the mature rate,cleavage rate and morula rate with the three mature media at high concentration of CO2. |
