| In this study, pepper Athelia rolfsii pathogen and 13 strains of Trichoderma which can antagonise the pathogen were isolated.The 13th strain of Trichoderma was filtered with confrontation test.The affection of culturing conditions were considered for the 13th strain of Trichoderma, The research results showed that the 13th strain of Trichoderma can patience heat and acidity. It can grow well when the culture temperatuer reach 50℃, In neutral or acidic environment,it can grow fast. In alkaline conditions, the growth of the 13th strain of Trichoderma can be inhibited.Also,the illumination is another factor to effect the growth of the 13th strain of Trich-oderma,the research result showed that there is no effects to the growth of the 13th strain of Trichoderma when it given strong sunlight for 6 hours. The 13th strain of Trichoderma grow fast, it is good for competing living space and nutritive material,which is good for antibiotic effects.Universal primer for identification of fungi ITS1 and ITS4 were used for PCR amplificating the ribosomal DNA ITS sequences of the 13th strain of Trichoderma. The ITS sequence alignment in Genbank showed that the sequence is the same with the accession number for the ITS sequence identical strains EU280124.1, indicating the 13th strain of Trichoderma is the same specie with the strain whose accesed number is EU280124.1:T.hamatum.For further study the inhibition of pathogenic mechanism of Trichoderma, the chitinase activity of the 13th strain of Trichoderma were measured of different fermentation time.The results show that chitinase activity reached maximum.At the same time we did the experiments on the electron microscopy,we use Trichoderma, pure chitinase and chitinase extracted by fermentation of crude enzyme extracted to treat Pepper Athelia rolfsii pathogen,the results show that the three treatments can degradation and destruction of peppers southern blight pathogen mycelium, mycelium surface degradation can make or break down, so that surface of fold-like hyphae, thus, so we can say that Trichoderma chitinase produced play a important role in biological control mechanism. |
