To investigate the developmental expression patterns of adiponectin (ADIPOQ) and adiponectin receptors 1 and 2 (ADIPOR1 and ADIPOR2)genes at the age of 7 months in fat and muscle tissues of Landrace (a leaner Western breed) and Rongchang pigs (a fatty indigenous Chinese breed), Real-time fluorescence quantitative PCR was applied. In addition, the genes expression difference between breeds, genders and tissues and the effects on adipose traits were analyzed.Results showed that Rongchang pigs had higher adipocyte deposit capability than Landrace pigs and lower lean percentage in terms of backfat thickness. intramuscular fat content and Eye muscle area. The studies indicated that Rongchang pigs had better meat quality than Landrace pigs by measuring fatty acid content and pH value in ten tissues of Landrace and Rongchang pigs respectively.The expression of adiponectin (ADIPOQ) and adiponectin receptors 1 and 2 (ADIPOR1 and ADIPOR2) mRNA in the adipose and muscle tissues exhibited the specific different patterns by genderes and breeds. The expression level of ADIPOQ mRNA in Landrace male pigs was significantly higher than in Landrace female pigs in perirenal fat, greater omentum, intramuscular fat and inner layer of back fat (P<0.01). At the same time it was significantly lower in pericardial fat and inner layer of back fat (P<0.01). The expression level of ADIPOQ mRNA in Rongchang male pigs was lower than the Rongchang female pigs in all tissues apart from longissimus muscle. Abdominal subcutaneous fat of which reached a very significant level (P<0.01). And the inner layer of back fat reached a significant level (P<0.05). The expression level of ADIPOR1 mRNA in Landrace male pigs was significantly higher than in Landrace female pigs in perirenal fat,abdominal subcutaneous fat, intramuscular fat, longissimus muscle and psoas(P<0.01). Meanwhile, it was significantly higher in inner layer of back fat (P<0.05);very significantly lower in lesser omentum (P<0.01); significantly lower in upper layer of back fat (P<0.05). The mRNA abundunce of ADIPOR1 mRNA in Rongchang male pigs was very significantly higher than in Rongchangfemale pigs in upper layer of back fat (P< 0.01). The expression level of ADIPOR2 mRNA in Landrace male pigs was significantly higher than in Landrace female pigs in perirenal fat and inner layer of back fat (P< 0.01).Meanwhile, it was significantly higher in abdominal subcutaneous fat (P<0.05); significantly lower in pericardial fat(P<0.05).The mRNA abundunce of ADIPOR2 mRNA in Rongchang male pigs was very significantly higher than in Rongchangfemale pigs in abdominal subcutaneous fat (P<0.01).At the same time it was very significantly lower in ntramuscular fat(P<0.01); significantly higher in pericardial fat (P<0.05).The expression level of ADIPOQ mRNA in Landrace female pigs was higher than in Rongchang female pigs in all ten tissues. And there were no significant differences only in longissimus muscle, psoas and perirenal fat. The expression level of ADIPOQ mRNA in Landrace male pigs was higher than in Rongchang male pigs in all ten tissues.And there were no significant differences only in abdominal subcutaneous fat,upper layer of back fat,longissimus muscle and psoas. he expression level of ADIPOR1 mRNA in Landrace female pigs was very significantly higher than in Rongchang female pigs in upper layer of back fat(P<0.01), instead,very significantly lower in perirenal fat(P<0.01). The expression level of ADIPOR1 mRNA in Landrace male pigs was significantly higher than in Rongchang female pigs in perirenal fat,greater omentum, inner layer of back fat and pericardial fat (P<0.01). Meanwhile, it was significantly lower than in Rongchang male pigs in upper layer of back fat (P<0.01). The expression level of ADIPOR2 mRNA in Landrace female pigs was higher than the Rongchang female pigs in all tissues apart from intramuscular fat. There were quite significant differences in perirenal fat, greater omentum, abdominal subcutaneous fat, intramuscular fat, upper layer of back fat, inner layer of back fat and pericardial fat.The expression level of ADIPOR2 mRNA in Landrace male pigs was higher than in the Rongchang male pigs in all tissues. There were quite significant differences in perirenal fat,greater omentum, abdominal subcutaneous fat, intramuscular fat, inner layer of back fat and pericardial fat.So this experiment is benefit to discuss the molecular mechanisms of swine fatty deposition in different tissues and it offers the basic experiment facts to the theory.
