To study the action of cryptdin-1 and cryptdin-4 in the UC process,this experiment selected BALB/c mice as the object and let them drink freely 7% DSS solution for 7 days in order to make acute UC model. we took the mouse duodenum, jejunum, ileum and colon out after 1day,3days,5days and 7days, and then detected the cryptdin-1 and cryptdin-4 expression in the intestinal tract by real time quantitative PCR and the correlation between them.The main results are as follows:1.The levels of cryp-1 in the same segment of the intestinal tract in different phase of UC: In the small intestine, cryptdin-1 mRNA was in the highest level at the the 1-day and was highly significant difference with the other 3 periods(P<0.01).The second volume expression was at the 5-day with the 3-day level significantly difference(P< 0.05). The expression of the 7-day was the lowest and was significantly inferior to the other 3 periods(P<0.01). In the colon, the expression was at highest level in the 1day and also significantly higher than the other 3 periods(P<0.01).The expression is poorer at the 7-day. The minimum amount of relative expression was at the 5-day.2.The cryp-1 mRNA expression at the same period of UC model but in the different intestinal tract parts: At the 1-day,cryptdin-1 mRNA was the highest expression in duodenal ,significantly with other three bowel segments(P<0.05). Jejunal expression took the second place. In the colon ,the level was the minimum and was highly significant difference with the other three segments(P<0.01). At the 3-day and the 5-day, the duodenal level was the highest. Colon,s expression was significantly below than the duodenum, jejunum, ileum expression level (P<0.01). When the 7-day , the highest expression was in jejunum, then was in the ileum, the lowest was in the colon, and three parts were extremely significant difference among each other (P< 0.01).3. The levels of cryp-4 in the same segment of the intestinal tract in different phase of UC: In the duodenum and jejunum,the level was reached the highest amount at the 1-day,and was significantly higher than the7-day and the 3-day(P<0.01).The second expression level was at the 5 day. The 7-day volume in the expression was minimal.In the ileum, the cryp-4mRNA was the highest in the 5 day,with significantly higher than the other periods(P<0.05). Second, the 1day level was compared with the 3-day and the 7-day was highly significant(P< 0.01). The lowest level at the 7-day. In the colon, cryptdin-4 was the highest level at the 7-day, with the three periods differences significantly(P< 0.01). The 3-day levels were at the lowest.4.The levels of cryp-4 in the different segment of the intestinal tract in the same phases of UC: At the 1-day and 3-day, cryptdin-4 mRNA was the highest in the colon and was extremely significant difference with duodenum's(P<0.01). The colon expression was at the second level,with significant difference(P<0.05). The colon was lowest. The 5-day, duodenal expression with other parts was the highest and was extremely significant(P<0.01). And the ileum's level took the second place(P<0.05). Also in the colon ,the expression was the lowest. The 7-day, the colon level was the highest and was higher significantly than the duodenum(P<0.01). Minimum of three parts was the duodenal level, extremely significant difference among the three(P< 0.01).5. The correlation between cryptdin-1 and cryptdin-4 in the acute UC model :The cryptdin-1 and cryptdin-4 mRNA expression in the process of UC model was a very significant positive correlation (r = 0.801, P<0.01).These results suggest that cryptdin-1 and cryptdin-4 both expressed in UC and its expression had significantly correlation. In the UC,cryptdin-1and cryptdin-4 are in the highest amount in the duodenum and jejunum at the 1-day .The 7-day's expression reached to a minimum level. In the colon,cryptdin-1 and cryptdin-4 expressed at low leval, cryptdin-1 is the highest expression at the 1-day,while cryptdin-4 is the7-day. It showed that cryptdin-1and-4 had expression in the whole process of UC model, having the close relationship with UC and playing an important role.
