The colitis model chicks were used in this study. The modern molecular biology techniques combined with traditional pathology testing methods were used for discovering the changes of the number of white blood cells in blood and dendritic cells in cecal tonsil, the expression of TNF-α、 TLR-4、IL-10mRNA in colon, the clinical signs and pathological changes. The study aimed at artificially copying the colitis with2,4,6-trinitrobenzene sulfonic acid solution mixed with50%ethanol (TNBS ethanol mixture) and showing the inhibition of inflammation and influence of innate immunity in colitis chicks after the application of probiotics. The results showed that:1. TNBS ethanol mixture can be given through the gut of chicks successfully replicated in animal models of ulcerative colitis.2. It is3to8days after chicks was given TNBS ethanol mixture through the gut of them. The number of leukocytes, eosinophils and basophils in their peripheral blood is different degrees higher than the control chicks (P<0.05or P<0.01), but it is significantly or very significantly (P <0.05or P<0.01) higher than the control chicks at the4day.3. It is3to8days after chicks was given TNBS ethanol mixture through the gut of them. The expression levels of TNF-α, TLR-4, IL-10mRNA in their colon tissue are various degrees higher than the control chicks (P<0.05or P<0.01). Those results indicated that TNBS ethanol mixture could significantly promote the differentiation and maturation of chicks’ white blood cells in their peripheral blood and it also could synthetic and release a large number of pro-inflammatory cytokines. So it could expansion the role of inflammation in the damage to the body.4. It is3to8days after chicks was given TNBS ethanol mixture through the gut of them. The number of leukocytes, eosinophils and basophils in their peripheral blood is significantly or very significantly (P<0.05or P<0.01) higher than the ulcerative colitis chicks intervened by the application of probiotics, but it is significantly or very significantly (P<0.05or P<0.01) lower than them at the4day.5. It is3to8days after chicks was given TNBS ethanol mixture through the gut of them. The expression levels of TNF-α and TLR-4mRNA in their colon tissue are significantly or very significantly (P<0.05or P<0.01) higher than the ulcerative colitis chicks intervened by the application of probiotics, but IL-10mRNA expression level is significantly or very significantly (P<0,05or P<0.01) lower than the ulcerative colitis chicks intervened by the application of probiotics. Those results indicated that probiotics not only could upregulate the expression of anti-inflammatory cytokines gene, but also downregulate the expression of pro-inflammatory cytokines gene. So it can suppress the occurrence and development of inflammation to achieve remission of inflammation symptoms and reduce inflammation the damage to the body.6. It is3to14days after chicks was given TNBS ethanol mixture through the gut of them. The number of dendritic cells in their cecal tonsil were significantly or very significantly (P<0.05or P<0.01) lower than ulcerative colitis chicks intervened by the application of probiotics and control chicks.7. It is3to14days after chicks was given TNBS ethanol mixture through the gut of them. Their body weight were different levels lower than the control chicks and the ulcerative colitis chicks intervened by the application of probiotics (P<0.05or P<0.01).
