Anji Baicha is a natural mutant tea plant (Camellia sinensis), which was discovered in the Zhejiang province. Its leaves has stage albescent phenomenon. During stage albescent process, the content of amino acid in white leaves is 2-3 times higher than what in green leaves. This paper used cDNA-AFLP technology isolation of differentially expressed genes during stage albescent process of Anji Baicha, and analysed sequences of differentially expressed genes. Done this for find excellent quality-related genes so as to do make sense to full develop and utilize our country tea resources. The main research results can be listed as follows:1.The cDNA-AFLP system of high resolution and reproduction was established by sliver staining technology. According to the established cDNA-AFLP silver staining technique system to study on Anji Baicha, the same or different primers combination amplify albescent or green period leaves was quiet abandunt.2. Gene expression of Anji Baicha albescent and green leaves was analyzed by cDNA-AFLP technology. The result shows that there were apparent differential expressions.163 fragments spcially display in albescent period while 62 fragments spcially display in green period.135 fragments of better reproduction were cloned, sequenced and BlastX. Sequence analysis indicated that among 31 squenced fragments have homologies,5 sequences were separately related to cold, drought, catechins metabolism, respiration, water transport channel.18 sequences related to cell cycle,4 sequences related to signal transduction,3 sequences related to Alkaloid synthesis,1 sequence unknown its function, and no homologies were found in Genebank for the other 104 fragments.3.To verify expression level of the 31 out of homology of Anji Baicha albescent and green leaves by RT-PCR technique. The results showed the expression level of 7 genes were higher in white period. The expressed products of differential genes were probable related to some different characters like high amino acid or pigment changes. Through the study, the cDNA-AFLP system was established suit for tea plant, and differential expressed genes were isolated during albescent period. From what study on, it can settle foundment of further research Anji Baicha quality differences formation molecular mechanism during albescent period.
