The experiment began in May 2008. Using the stem with bud of bitter gourd "bixiu" as explants. By means of analysis of variance and multiple comparisons about sampling time, disinfectors and sterilization time, preincubate, hormones we establish the system of stem with bud in vitro culture. The results were as follows:(1)May was proved to be better as the sampling time because the contamination rate is lower than that in June to August.(2)0.1%HgCl2 was the better than 2%NaCl.O.6min was the optimal surface sterilization time.(3)Preincubate reduced either the contamination rate or the differentiation rate.(4)6-BA and IB A have great impact on differentiation, but IAA and NAA failed to.MS+6-BA4.0 mg·L-1+0.2 mg·L-1 was the best medium for differentiation in which the rate of differentiation was 43.33%. Higher or Lower concentration of 6-BA were not conducive to the differentiation.(5)Adding AgNO3 to the proliferation medium improved the regeneration frequency, but inhibited the plantlet growth; the optimal concentration was 1.0mg·L-1(6)Adding GA3 improved the plantlet growth,but the plantlet had excessive growth when the concentration of GA3 was 3 mg·L(7)6-BA4.0 mg·L-1+IBA0.2mg·L-1+AgNO3 1 mg·L-1 +GA32 mg·L-1 was the best proliferation medium in which the proliferation confficient was 4.75. (8)The plantlet could not grow when just added IBA to the medium without 6-BA.(9)AgNO3 was not conducive to radication. The optimal proliferation medium for shoot from stem was:1/2MS+20G/Lsucrose+0.5mg·L-1KT+0.4mg·L-1IBA.(10)The explants had high differentiation rate and fine growth when IAA/ZT=1.6. The explants had excessive differentiation when the ratio is lower and little when the ratio is higher.
