Brucellosis is a chronic infectious disease of humans. livestocks and wild lives by intracellular bacteria of the genus Brucella .Brucella, a member of the bacterial genus Brucella, is pathogen of the disease, which are small, gram-negitive, facultative intracellular bacteria. Brucella invad into the body mainly through the digestive tract, skin and respiratory mucosa and led to infection and morbidity. Most of Brucella parasitize mainly in lymphocyte and lympbatic tissue, resulting serious lesion of the immune system, followed by bacteriemia and toxaemia, which impair to nervous system , reproductive system and skeleton system..Brucellosis has important public health significance and huge economic losses for world.B. suis strain 2, B. melitensis strain 19 and B. melitensis strain M5-90 have been widely used in China as vaccine, which achived a great success in brucella control for animals for its high efficacy. However, classical serological diagnosis of brucellosis could discriminate vaccinal antibody from antibody induced by infection. Therefore, study of the antibody variation from vaccinated cattle and ovine using different methods is great value to diagnose brucellosis.In this sdudy, the vaccinated cattle with B. suis strain 2, B. melitensis strain 19 and B. melitensis strain M5-90 were bled prior to vaccination (0 day) and thereafter at 1, 2, 3, 4, 5, and 6 month post vaccination.The vaccinated ovine withB. melitensis strain M5-90 and B. suis strain 2 were bled prior to vaccination (0 day) and thereafter at 1,2,3,4,5 and 6 month post vaccination.Our results showed that the majority of animals showed positive reaction by RBPT,SAT,HVRI-cELISA and Svanova-cELISA 1 month post vaccination with B. suis strain 2, B. melitensis strain 19 and B. melitensis strain M5-90. the majority of animals showed positive reaction by Idexx-iELISA 1 month post vaccination with B. suis strain 2, and later 1month post vaccination with S19 and M5-90. The serum samples from cattle vaccinated with S2 post 6 months can not be detected positive reaction by HVRI–cELISA and Svanova-cELISA. The serum samples from cattle vaccinated with S19 post 6 months can not be detected positive reaction by Svanova-cELISA and Idexx-iELISA. The serum samples from cattle vaccinated with M5-90 post 6 months can not be detected positive reaction by all of tests, and lasting longer time.Studies have shown that the agreement rate between HVRI-cELISA and Svanova-cELISA when detecting sera from cattle S19 are the highest (kappa=0.927) than with other methods, and lower when detecting sera from cattle vaccinated with S2 and M5-90, but the kappa values are also more than 0.75 with a very good coincidence rate.The results of ovine vaccinated with M-90 and S2 showed the majority of animals showed positive reaction by RBPT,SAT,HVRI-cELISA and Svanova-cELISA 1 month post vaccination.The results obtained with HVRI-cELISA at 32% inhibition of the conjugate control and Svanova-cELISA was not significant (P>0.05). The serum samples from ovine vaccinated with S2 and M5-90 post 6 months can not be detected positive reaction by HVRI–cELISA at 32% inhibition of the conjugate control.
