Objective:To study the feasibility of repair of peripheral nerve defects with transplantation of embryonic spinal cord(ESC). Motbeds:36 rabbits were randomly divided into 3 groups. Two sides sciatic nerves were created defects of 8mm~ 16mm and 32mm(4 times~ 8 times and 16 times of diameter) by surgery The left defects were bridged by ESC of rabbit that the gestational age was 25?8 days The right defects were bndged with autogenous sctatic nerves that were excised from left The preoperative and postoperative contents of IL-2, IL-6, lgA, IgG and 1gM in blood were determined. The motor nerve conduction velocity (MNCV), latency(Lat) and aniplitude(Amp) of muscle tibiahs anterior were examined by electromyograph at the first ,the second and the third month end After electromyogram checking, the animals of each groups were killed and the specimens were sampled,fixed, sectioned and stained,then to observat the conditions of neuranagenesis by light microscope and electron microscope The two sides muscle tibialis anterior of all rabbits were extracted and their wet weight were measured The tracing of reverse by fluoro- gold(FG) and horseradish peroxidase (HRP) was used at experimental sides in parts animals of every groups, for imderstandrng the conditions of their nerve iter. Results: There was no significant difference between the peroperative and postoperative contents of IL-2,IL-6, IgA, lgG and IgM(p>O.05).This may be explained it without significant reaction of immunity and rejection after transplantation of ESC .The postoperative contents of MNCV~ Lat and Amp of two sides showed no obvious difference (P>O.05).The nerve fibers of embryonic spinal cord transplanted and their distal nerves were coming on well, the quantity of medullated nerve fiber of unit area of two sides showed no evident discrepancy (P>O.05). There are a large number of ripe nerve fibers in the transplanted parts of embryonic spinal cord, of all these normal Schwann抯 cells, the integrated basilemma, the clear and discernable Iamella of myelin sheath were observed by electro microscope.The neurofilaments and microtubules of axial fibers were well distribution.The diameter of medullated nerve fibers and the thickness of myelin sheaths of the two sides showed no obvious difference(P>O.05),dealt with by compute picture analyse sistem.There are no apparent diversity between the left and right wet weight of muscle tibialis anterior (P>O.05)The nerve-cells of dyed were discovered at ventricornu after the tracing of reverse by FG andl-IRP . Conclusion: It is feasible that repair of peripheral nerve defects with transplantation of embryonic spinal cord in this experiment.
