OBJECTIVE: Our purpose was to investigate the expression of Leukaemia inhibitory factor in human endometrium throughout the menstrual cycle. The difTereflt expression of LIE in endometriutn between fertile women with unexplained infertility woman in the middle and late secretaiy phase. Modulate of LIE expression by estradiol and progesterone and TNF- a and IFN- i. METHODS: Normal endon~trium tissue samples were obtained from 6 patients to put up uterus conceptive thingF 26 patients in Nanfang hospital reproductive center undergoing in vitro fertilization-embryo transfer (IVE-ET), 3 patients with ovary cy~ic and 3 patients with uterine myopia. Some endometrium was fixed in 10% buffered formalin (PH 7.4) for immunohistochemistry and other samples was put into D-Hank抯 to experiment for cell culture. By a specific and sensitive enzyme-linked -4- immunosorbent assay (ELISA) the supernants were examined and put estradiol , progesterone and TNF- a and LFN- Y for 24h the supernants were examined any more. RESULTS: I.. We demonstrated that positive immunostaining for LIF was observed in all human endometrial tissues ~ A moderate level of immunoreactivity was observed in endometrial stroma throughout the menstrual cycle. Immunostaining in luminal and glandular epithelium varied throughout the cycle. There are no immunostaining in the proliferative and in the early secretary phases immunostaining was relatively low, while strong immunostaining intensity occurred in the middle secretary phase.2.. The in vitro endometrial LW secretion by cell cultures from fertile women were higher than that of unexplained infertility women. (541.2? 158.6pg vs. 139.9 ?9.Spg, P 0.05). CONCLUSION: It was suggested that: 1.. LIF may play a important role in embryo implantation. 2 . Lower expression of LIF in endometrium on middle secretary phase may be responsible for -5- unexplained infertility. 3. Progesterone and TNF- ~a can participate in embryo implantation by increasing the mount of LIF in endometrinni, [FN- i inhibit the expression of LLF may be one of the cause of inWlantation failure.
