AIM: Though extensively investigated, the etiology of endometriosis has remained enigmatic. The retrograde menstruation and implantation of endometrial fragments is a primary mode of developing endometriosis in the peritoneal cavity, but this phenomenon is nearly universal. Some reports have stated that an immunologic alteration plays a role in the development and progression of endometriosis. Our purpose is to determine the effect of angiogenesis growth factors(IL-8, PDGF, VEGF), immunomodulator(IFN- a -2b, IFN- Y ) and ECM (collagen I > III) on developing of endometriosis. METHODS: 1. Eutopic endometrial cells from with endometriosis women were cultured and purified, identified. Collected epithelia cells were treated with various concentrations of IL-8, PDGF, IFN- a -2b, IFN- Y in 96 well plate, then were observed its effect through MTT method. Epithelia cells and stromal cells were treated with Ing/ml concentration of IL-8 then observed by transmission electron mircoscopy. Part of epithelia cells were treated with lOng/ml concentration of PDGF then measured the stained areas of VEGF. 2. Normal eutopic endometrium(group C), red lesion (group A) and black lesion (group B) tissues were collected and type K III collagen was detectived by sinus red and polarization microscopy. The percentage of the stained area in the tissue was measured with a computerized image analysis system.(POHOTOSHOP) . Result: 1. IL-8 and PDGF were found to stimulate proliferation of endometrial cells and were notable with lOng/ml (P<0.05f<0.05). Endometrial cells has high secretory activity .Collagen has been produced by endometrial cells. Epithelial cells can be detected VEGF in with PDGF and with_no PDGF. and the stained areas of VEGF in with PDGF is higher than that of control (P<0.05) . IFN-a -2b and IFN-Y was found to restrain proliferation of epithelial cells, there were significantly difference in the OD value of those groups which were treated with IFN-a -2b (50IU/ml) and IFN-Y (100IU/ml)than in the control group (P<0.05, P<0.05). 2. There were significant difference among the three groups. The lowest concentrate of type I, III collagen is group C (P<0.05,P<0.05). Then the concentrate of type III collagen in group A is the highest among the groups (P<0.05). Group B is the highest concentrate of type collagenl (P<0.05). Conclusion: IL-8, PDGF and VEGF are positive angiogenesis growth factors. They participate in pathogenesis of endometriosis by autocrine and paracrine. Abnormal concentration of I, III type collagen may induce abnormal immunoreaction in endometriosis. Both IFN- a -2b and IFN- Y are immunostimulative factors. They can modify the immune system in patients with endometriosis. Immunostimulative therapy is a new method of curing endometriosis.
