| Objective To investigate the effects of Prostaglandin E1 (PGEI) and recombinant human growth hormone (rhGH) for liver ischaemia-reperfusion injury and liver regeneration in rabbit model. Methods 48 rabbits were divided into four groups randomly. In this study, hepatoduodenal ligament was occluded for 45 mm to induce the liver ischaemia. The left lateral lobe hepatectomy was performed after the ischaemia was present for 40mm. In the group A (control group, n=12), normal saline was used to instead of the PGE1 or rhGH. In the group B (PGE, group, n12), PGE1was infused continuously through auricle vein by pump with a rate of 0.5 i-i g . kg~1 . mm?for a 10mm preischaemic period and a 120-mm post-ischaemic reperfusion period. In the group C (rhGH group, n=12), rhGH was injected hyodermicly in the back of ear with the dose of lU/kg qdX7day. In the group D (PGEI and rhGH combined group, n~l2), PGE1 and rhGH was used simultaneously as same as group A and group B. Serum biochemical analysis (alanine aminotransferase: ALT, aspartate aminotransferase: AST, lactate dehydrogenase: LDH, superoxide dismutase: SOD and molondialdehyde: MDA) was carried out at preischaemic, ischaemic for 40 mm, reperfusion for 1 hr, 24hrs, 72hrs and l4days. Proliferating cell nuclear antigen label index (PCNA-LI) was detected at preischaemic, 72hrs and l4days in liver tissue. Moreover, histological examination was carried out at preischaemic, ischaemic for 40 mm, reperfusion for 1 hr, 72hrs and l4days respectively. The change of rabbit weight in 3days, l4days and the mortalitywithin 3days after operation was analysized.
