Objective: To improve the clinical therapy, studies were performed on the details of Th-cell polarization, intracellular IFN?y secretion and CD69 expression on NK cell in vitro for investigating the statute of cellular immunity of the patients with different courses of condyloma acuminate(CA). Methods: After isolated the PBMC and blood-plasma from anti-clotting human peripheral blood with heparin by Ficoll-Hypaque centrifugation, the PBMC culture procedure in vitro and two-color immunofluorescent staining were undertaken as following: (1) PBMC were polarized 72 hours by PHA (2O~g/mL) and the self-bloodlasma(BP) together, then followed by two-color immunofluorescent staining with anti-CD4-PE and anti-CCR5-FITC or anti-CD4-FITC and anti-CCR3-PE . (2) PBMC were activated 2,12,24 hours by PDB (iO-~ molIL) ,then stained with anti-CD56-FITC and anti-CD69-PE. (3) PBMC were stimulated 4 hours by PDB (1 O4 molIL) , Ionomycin (1 mglmL) and Monensin (2mM) together, then intracellular stained with anti-CD56-PE and anti- IFN- Y -FITC. At last the stained cells were analyzed by flow cytometry. Results: 1. Th-cell polarization: The percentages of Th1/Tb2 cells of control group were (16.68±1.56) %/( 16.46ad.12) %, and the ratios ofThl/Th2 cells were 1. 01 ±0. 05. The percentages of Thl/Th2 cells of the short-history-patients with CA(SH-CA) group were (25. 82±2. 22) %/ (14. 80±1. 14) % , the difference between them and those of normal group was significant (P<0.05) ,and the ratios ofThl/Th2cellsofSH-CAgroupwere 1.15?.11, which showcdThl response; ThepercentagesofLH-CAgroupwere (12. 20?. 37) %/ (13. 74?. 99) %, the difference between them and those of normal group was significant (P<0.05) ,and the ratios of Th1/Tb2 cells was 0.89 11, which showed Th2 type response. 2. NK-cell activation: The percentages of NK-cells expressing CD69 of control group at 2,12,24 hour were (62. 99?. 62) %, ( 80. 97?. 16) %, (91. 26?. 83) % respectively. The percentages of the SH-CA group were (62.22?.01) %, (80.67?.20) %, (90. 30?. 75) %respectively, the differences between them and those of control group were no significant (P0.05) The percentages of the LH-CA group were (25.73?. 99) %, (57. 08?. 74) %, (67. 07 ?2. 52) % respectively, the differences between them and those of control group were significant (P<0.05) . The ratios of IFN- i positive NK-cells control group and SH-CA group were (61.62?.65) % , (61.91?.48) % respectively, the difference between them was no significant (P>0.05) . The ratios of LH-CA group were (34.69?.36)% , the difference between them and control group was significant (P<0.05) Conclusions: There were Thl type responses in the patients with SH-CA, but LH-CA showed Thl type response have switched to Th2 type response. The activation and function of NK cells the patients with SH-CA were normal, but those of the patients of LH-CA were deficient. These data would provide the theory and new ways for the immune therapy of CA, especially LH-CH.
