| [Objective]: To investigate the roles of COX-2 and VEGF in cell proliferation and angiogenesis of condyloma acuminate (CA). [Methods]: The expressions of COX-2 and VEGF proteins and microvessel density (MVD) in 40 cases of CA and 15 normal prepuce skins were examined with immunohistochemistry staining.Furthermore; we analyzed the relationships between the expressions of these markers. We also examined the expressions of COX-2 and VEGF mRNA using RT-PCR method, in the 12 fresh cases of CA and 5 normal skin tissues.[Results]: The MVD of CA was much higher than that of normal control (P<0.05) . The positive expression rates of COX-2 and VEGF in CA were 62.5% and 92.5% respectively ,which were significantly higher than that of normal controls (P<0.05 ). The distribution of COX-2 and VEGF was throughout the epidermis, mostly were strong positive, while there was only weak expression in basal layer in normal controls ;In severe cases ,the expressions of COX-2,VEGF were stronger than that of light cases (P<0.05) ;There were no close correlation between the expressions of COX-2,VEGF and the course of disease(P>0.05) . The MVD in COX-2 positive cases were higher than that of negative cases.Furthermore, the expression of COX-2 was closely correlated with that of VEGF (P<0.05) ,and MVD also had close correlation with COX-2,VEGF. The positive expression rate of COX-2 mRNA was higher than that of normal control (P<0.05) .There was no significant difference of VEGFmRNA expression between CA and normal control(P>0.05),but the expression level in CA is higher than in normal control (P<0.05) .[Conclusions]: COX-2 and VEGF promoted the pathogenesis of CA together by promoting the growth of cells and microvessel. The stronger expressions of the two in severe cases accelerated CA growth further. There were no distinction expressions of the two between different disease courses. It may be an important method of CA therapy to suppress the expression of COX-2 and VEGF. |
