Background:Interleukin 12(IL-12) and interleukin 10(IL-10) both are important to the induction and specialization of Th cells, which are the key to control the immunological reaction in viral hepatitis B.But the in situ study about IL-12 or IL-10 in human liver has not ever been reported. Here, the role of IL-12 and IL-b in viral hepatitis B were studied in liver tissue.Methods:IL-12 and IL-10 were detected with ELISA in serum and semi-quantatitive histoimmunochemistry in liver tissue slices in 45 viral hepatitis B cases of whom 29 cases received interferon- ci treatment. Results: IL- 12 and IL- 10 both exist mostly in liver cell cytoplasm and on liver cell membrane. The analysis showed positive correlation between IL- 12 and IL- 10 as well as IL-10 and histological activity grade(G) and negative correlation between I-lIBcAg in liver and the ratio of IL- 12 to IL- 10. The quantity of IL- 12 and IL- 10 in those positively responding to IFN-α are both positively higher than in those who had negative responses to IFN-α .Conclusion:IL-12 is important to the execution of I-IBV;IL-10 reflects the immunological activity in liver;the assay of IL-12 and IL-b is useful to prospect the effect of IFN-α...
