Objective: To preliminarily investigated varities of cell cycle,apoptosis of cells and contents of nitric oxide (NO) and endothelin 1 (ET- 1) in culture solution of primary HIUVECs by radiation and its relationship with dose-response and time course. Method: HIUVECs was primary cultured, then identified with transmission electron microscope(TEM) and Immunohistochemical stains of antibody of human VIII factor. After growth to confluent, HIUVECs was irradiated with 6MV X-ray; then detected varities of cell cycle with flow cytometry(FCM); Observed and detected apoptosis of radiated I-lU VECs with TEM and FCM determining contents of Er-i and NO by radioimmunoassay and HINO3 reductase method respectively. Result: The cultured primary I-IUVECs growthed very well, we found W-P body with TEM and positive expression antigen of factor VIII in the I-IUVECs and been proved endothelial cells. After irradiated with 0 ~?2OGy, detected decrease in GJ/G1 phase and increase in G2/M phase of radiated confluent H7UVECs ~vith 5 ~- 2OGy through assay of FCM at 24 and 48 hour, but not changed after radiated with 2Gy. TEM and FCM failed to detected increase in apoptosis and death of primary HIUVECs induced by 0- lOGy X-ray at 24 or 48 hour. ET-l contents of culture solution of HUVECs that shamed radiation was 1304.47?126.86 pg/i X 10cells/ml during 24 hour, we found decrease after 520Gy radiation, P<0.05; N02 contents of culture solution is 48.39?2.49 .1 mol/lxlO5cells/mI in HJJVECs shamed radiation during 24 hour, radiated with 0?0Gy X-ray unchanged it. Conclusion: The cultured primav HUVECs was proved endothelial cells and could be used in relative study. Apoptosis and death of confluent primary l-LUVECs wasn't increased during 48hour after 0 lOGy radiation; radiation with O-2OGy unchanged NO contents secreted from I-IIJVECs. Only HUVECs was radiated with a single bigger dose(~5Gy) could induce retardation in stage G2M and decrease ET- 1 contents of culture solution.
