Objective:The present study was performed to invesitage the effects of Experimental hyperlipidemia serum and Zhengzhi Pill (ZZP)-containing serum on the productions of nitric oxide (NO) and endothelin(ET) by endothelial cells. The injury effect of experimental hyperlipidemia serum as well as the lipid-modulation effect,endothelial protective effect and anti-atherosclerosis effect of ZZP was explored to elucidate the pharmacological mechanism of ZZP. The practical application of serum lipoprotein electrophoretagram(SLEG) in detecting the abnormal lipoprotein was also discussed.Methods:Experimental hyperlipidemia serum and Zhengzhi Pill (ZZP)-containing serum were prepared respectively. Endothelial cells were cultured with both serum and the levels of NO and ET in supernatants were detected. The sera obtained from animals treated with normal diet or high fat diet were analyzed by both routin methods and SLEGResult:Experimental hyperlipidemia serum significantly inhibited NO production and stimulated ET expression (P<0.01). ZZP showed a dose-dependent effect of increasing NO production and simultaneously inhibited ET expression (P<0.01). SLEG is more sensitive than routine detective method in reveal the lipoprotein abnormality. The serum,which is detected normaly by routine method but abnormally by SLEG,also showed the effect of inhibiting NO production and stimulating Et expression. Conclusion:1. Experimental hyperlipidemia serum damages the endothelial cells and breaks the balance between NO and ET. 2. ZZP has atherosclerosis protective effect. The mechanism maybe relate to its endothelial protective effect of regulating the balance between NO and ET. 3. SLEG is more sensitive in reflecting the liporpein abnormality.
