Lung cancer is a malignant disease that is extremely harmful to the life and health of human being. Its genesis is related to multiple faors. It has been proved there are proximately 70% of lung cancer with p53 gene abnormality based on the wide genetic and molecular investigation. p73, a new gene, has been identilied as a p53 homologue for their similarity of structure and function. It inhabits cell growth and induces cell apoptosis. Although p73 has a striking similarity to p53, there are some significant differences between these two genes. p73 gene mRNA is over-expression in some limited cancers and its silent allele is activated in some tumors. However the real function of p73 in tumongenesis is unclear and needs more studies. To elucidate the role of p73 in tumor development and to find a new clue of the tumor diagnosis and treatment, the expression of p73 gene was investigated in non-small cell lung carcinoma. A total of 46 specimens of non-small cell carcinoma and matched para-oancer tissue were screened for the levels of p73 mRNA using RT-PCR and analyzed the alteration of p73 by PCR and SSCP. The results showed that there were 24 lung cancer tissues with higher level expression and only 2 paired para-cancer specimens with higher-level p73mRNA expression. The expression of p73 mRNA in lung tumors was distributed significantly higher than that in para-cancer. (p0.01). The expression of p73 mRNA in low differentiated lung tumor was distributed significantly higher than that in high differentiation (P<0.05), but there was no significant difference between clinical I II and ll-IV (P>0.05)0 The result of allele- specific analysis showed that 13 of 15 heterozygous lung cancer specimens were with expression of G/C: NT and the paired pars-cancer specimens were with expression of GIC. No mutation was detected by PCR- SSCP. Based on the findings of this study, we can draw the following conclusions: (Dp73mRNA is associated with non-small cell lung cancer's tumorigenesis() p73mRNA may be closely related to differentiate levels of lung cancer but not related to its histological classification and clinical stages.The p73 silent allele in para-cancer tissues is activated in tumors, and it may be involved in the lung cancer's tumorigenesis.(J Mutation is not the main way that p73 takes part in the lung carcinoma's development Xjp73 may not be a suppressor gene.
