Surgical resection remains the first choice for curative treatment of hepatocellular carcinoma (HCC),but it was limited only to a small proportion of patients who have so-called small HCC without extrahepatic metastasis and with the tumour located away from major vessels.The survival period of patients who had no treated were 3 6 months.For the majority of patients,non-surgical treatment is the only alternative.Transcatheter hepatic arterial embolization (TAE)has been shown to prolong the survival period of patients and improve patients iving quality,and has been a primary method for advanced stage patients.but because the inducible overexpression of the multidrug gene and the function abnormity of P53 gene,the curative effects of TAE were decreased after long-term therapy.Heat(42 C -45 C ) can enhance the sensitivity of tumor cells to drugs,so it could be a new effective way to enhance the curative effects of TAE. Interventional radiology advanced rapidly in recent years,making the conceive of interventional heated chemotherapy to come true. Jnterventional heated chemotherapy combined the advantages of thermotherapy,chemotherapy and embolotherapy~and could become a new effective method to solve the heating problem of deep tumors. Objective: To observe the curative effects of treatment and in vivo study the mechanism of hyperthermal perfusion chemotherapy in VX2 rabbits to find a more effective method to cure HCC, and make experiment basis for the clinic application of Interventional heated chemotherapy. Methods: Vx-2 carcinoma cells were surgically implanted into the left liver lob of 60 New Zealand white rabbits,which were divided into 4 groups at random.with 1 rabbits for each group.To inject adriamycin(37Cor 600C),physiological saline(370Cc 60 0C )in different groups of VX2 tumour-bearing rabbits via hepatic artery. To observ the volume of tumour,the level of rabbits iver function,the survival period an photographs of pathological tissue. Results: l.After therapy,the tumour's growth rate of the group of adriamycin(60X2)wa significantly lower than the other treated groups(P<0.05),being significantly lower thai the untreated control group(P<0.01).Observed by ultrasound and compute tomography,the tumour's volume of the group of adriamycin(60)was significantl lower than the other treated groups(P<0.05).The survival period of adriamycin(60C group was significantly higher than the untreated control group (P<0.05). 2.After heated chemothrapy,The serum level of ALT and AST elevate significantly(P<0.05), but the restored in a week. No significant changes was fouru about y -GT,T-BJL,and ALB(P>0.05). 3.After heated chemothrapy,the photographs of pathological tissue showed that lot of tumor tissue were necrosed the fibrous tissue were proliferated,the vessels tumor were gored and obturated. some liver cells around tumor showe turbidity,swelling with infiltration of neutrophil leukocyte but recovered in a week.Conclusion: Adriamycin at 600C through hepatic artery perfusion can obviousl inhibit the growth of hepatic VX2 tumor,and prolong the survival period of tumor bearing rabbits,caused reversible injury on liver,and influenced the breath hear beat temperature,live function,but these side effects were slight.this result is of grea value for the clinical application of interventional heated chemotherapy.
