Objectives: The purpose of this study is to appraise the clinical effect of 1 O-Hydroxycamptothecin (HCPT) chemotherapy to human breast cancer n~ ~>~il and gene level throngL obscr\ ily 11w influrence of HCPT chemotherapy on Apoptosis Index of Human Breast Cancer cell Bcap-37, expression of bcl-2/bax gene. This study will be able to provide the reliable theoretical kas~ for es~ahliThine ~ind pati~iitw tIm treatment model for breast cancer. This study will also help to search for valuable tumor cyto parameters to appraise various chemotherapy effect for human breast cancer, and design individual treatment scheme according to the individually different biotypes. Thus this study will finally help to increase the survival rate of breast cancer patient especially prolong the period of disease free Methods: Experimental subjects: Human Breast Bcap-37 Cell line Experiment group: HCF chemotherapy, using HCPT, CTX, and 5-Fu. Try three different concentrations with the highest of HCPT 0.005mg/mi, CTX 0.1 mg/mi; 5-Fu 0.2 mg/mI. Then dilute them 4, 10 times to work as the middle and lowest concentrations. Control Group 1: CAF chemotherapy, using CTX, ADM and 5-Vu. Try three different concentrations with the highest of ADM 0.005mg/mi, CTX 0.1 mg/mi; 5-Fu 0.2 mg/mi. Then dilute them as the same times as above. Control Group 2: CMIF chemotherapy, using CTX, MTX and 5-Fu. Try three different concentrations with the highest of MTX 0.01mg/mi, CTX 0.1 mg/mi; 5-Fu 0.2 mg/mi. Then dilute them as the same times as above. Control Group3: blank group, without any treatment Use the above medication group to treat human cancer Bcap 3? .1! 1i~.c for 24 ho抜rs. Collect the treated cells, and measure the Al (Apoptosis Index), bcl-2/bax gene (this gene is related to apoptosis) expression level, and detecting the structure of cancer cell usinr~ TIE l\拁in~ method Result: Al of HCF group is significantly higher than that of CAF group and CMF group (P<0.05); bcl-2 expression rate of HCF group is much lower than that of CMF and CAF group under all the three concentrations (P<0.05). Inside each group, bcl-2 expression level decreases with increased concentration. For all the three concentrations, CMF group shows a higher hax expression than 1-ICF group. hut not significantly; Bax expression level of CAF group is significantly lower than that of HCF and CMF groups. Inside each group, bax expression level increases with increase in concentration (P<0.05); CAF group has higher mortality rate in all the three concentrations than that of HCF group, but the two groups show significant difference only at the highest concentration level, for middle and lowest concentrations, no significant difference in mortality rate is observed.
