Objective :To study the effect of ulinastatin(UTI) and taxotere(TXT) on cyclooxygenase-2,prostaglandin E2,interleukin 10 expression, proliferation ,invasion,apoptosis in the primary breast cancer cell.Methods : The enzyme digestion was extracted from human breast cancer cells,were cultured and randomly divided into four groups:control group,UTI group,TXT group,UTI+TXT group.By MTT experiment, transwell chamber invasion assay and flow cytometry cell technique experimental observation of the proliferation, invasion and apoptosis in cell;by enzyme-linked immunosorbent assay(ELISA) detection of PGE2 concentration in each group; by semi-quantitative PCR and western blot detection of COX2,IL10 gene and protein expression changes.Results : compared with the control cells,UTI and TXT can inhibit the proliferation,invasion and promotion apoptosis of the primary breast cancer cell,more significant joint UTI and TXT.UTI and TXT alone or combined treatment, COX2,IL10 expression was significantly decreased compared with the control group,while also effectively reduce the secretion of PGE2 levels of cells.Conclusion UTI and TXT can inhibit the growth of breast cancer, and a synergistic effect between the two ,the mechanism may be associated with UTI and TXT reduced COX2,PGE2,IL10 related。 Objective:To investigate the expression and the inhibition of Ulinastatin and Taxotere on cyclooxygenase-2,NM23,CASPASE-3,interleukin 10 in nude mouse with primry human breast cancer.Methods:To implantate primry human breast cancer cell in the right armpit subcu- taneous fat pad of BALB/c-nu/nu female nude mice.There were fifty mice divided into four groups with random.There were physiological saline group,UTI group,TAX group,UTI+TAX group. They all started injecting into abdominal cavity at the twenty-one days. The volume of primary tumor was established by the twenty-one days after tumor cell inoculation. Expression of cyclooxygenase-2,prostaglandin E2,interleukin 10,was detected by immunity class.Results:compared with the control group,UTI could significantly delay the increase of tumor volume in nude mice,but did not reduce tumor,TXT and UTI+TXT could significantly delay the increase of tumor volume in nude mice and tumor volume can reduced,and the UTI+TXT stronger than TXT,while UTI,TXT and UTI+TXT can increase NM23,CASPASE-3 and decreased COX-2,IL-10 espression。 Conclusion:UTI and TXT can inhibit growth of breast cancer ,and the combined dffects of two better than any of them,which may be with UTI and TXT reduced cox-2,il10 espression。...
