Objective We explored the effect of over-sexhormone on normal larynx of adult rabbit in order to reveal the correlativity between sex hormone and the laryngeal disease and attempted to provide the histological foundation about sex hormone to laryngeal disease in clinical treated.Method Forty healthy adult rabbits ranging in age from five to six months and in weight from 1.75 to2.25kg were divided into three groups(half of male and female).The androgen treatment group sixteen rabbits were injected Testosterone propionate ,5mg/kg body weight/week. The estrogen treatment group sixteen rabbits were injected Estradiol benzoate 0.5mg/kg/every other day, The control group eight rabbits were given nothing. All rabbit were individual killed in 1.2.4.8 weeks.Larynges were removed and than stored in a 10% formaldehyde. The stored larynges were then dissected and measured laryngeal anatomic numeral value, tissue slice and hmatoxylin and eosin staining individual measured the thickness of laryngeal epithelial in order to observe the changes in laryngeal mucous membrane and histological structure after the treatment.Result Two rabbits were dropouted because of dead in midde of the study. l.The laryngeal cavity became bigger and voice cards became longer and thicken by the over-androgen tre-atment. The over-estogen intensified the female character and lead to female characteristic on male. 2. Hematoxylin and eosin staining confirmed the similar laryngeal histomorphology of human and rabbit.It was observd that the epithelial proliferation and slight keratinization and squamatization of larynx,moreover, ciliated epithelium excess hyperplasia in subglottic region by over-sexhormone treatment. 3.Under the laryngeal mucous membrane the treatment rabbits were observd interstitial edema and plasma cells, as well as observed lymphocyte lymphadenosis proliferation and formed follide,especially in Reinke laryer. Furthermore the lumen of capillary enlarged ,deformed and filled with red blood cell were observd .On the other hand, it occurred that hyperadenosis and exuberance of glandular secretion in same region .Conclusions 1. Sex hormon have obviously effet on laryngeal epithelium. 2.The over-androgen lead to the changes on adult laryngeal cavity and muscle fiber,whereas the estrogeon effect were not obviously. Sex hormon have no effect on laryngeal nerve-tract and nerve fiber. 3.The larynx is a sex hormonal target organ. Stratified squamous epithelium of the larynx is a hormonal sensitive tissue.Therefore,it can have correlation between sex hormonal metabolic disorder and the occurrence and development of the laryngeal disease which derived from stratified squamous epithelium of the larynx such as laryngeal carcinoma and laryn geal papillomatosis. 4.Our data show that estrogen plays an important role in laryngeal trophism and immune defense. Estrogen replacement therapy may provide pre-vention and tretment of dystrophic pathologies of the vocal cords in postmenopausal women. It must go further into the question that the influence of sex hormone on larynx.
