Objective:To study the expression of the hyaluronic acid (HA), collagen, and aquaporin(AQP) in the larynx of D-galactose induced aging guinea pig and aged guinea pig. And to discuss the possibility of D-galactose induced aging guinea pig used for presbyphonia and the role of AQPs during the aging course.Methods:Thirty-two guinea pigs (4months old) were divided into two groups: the control group and D-galactose induced aging group (model group), with16animals in each group. Sixteen aged guinea pigs (2years old) acted as aged group. The animals of the model group were given subcutaneously injection of D-galactose with300mg/kg per day for six weeks to induce subacute aging model. There was no medication in the control group and the aged group. The expression of the HA, collagen, AQP1-5were detected using Alcian, Masson trichrome and immunohistochemistry, respectively.Results:The expression level of HA in the larynx of the control group, aged group and model group were0.798351,0.694163and0.724869, respectively. While the expression level of collagen of three groups were0.632295,0.71147and0.768211, respectively. The expression level of HA of the control group was higher than that of the model group, the expression level of HA of the model group was higher than that of the aged group. The expression level of collagen of the control group was lower than that of the aged group, the expression level of collagen of the aged group was lower than that of the model group. AQP1was expressed on the pseudo multiple cilium columnar epithelium, glands of the lamina propria, endotheliocyte of the micro vessel under the epidermis. The expression level of AQP1of the control group was higher than that of the model group, the expression level of the model group was higher than that of the aged group. AQP2was expressed on the glands of the lamina propria mucosa, AQP3was expressed on the pseudo multiple cilium columnar epithelium. The expression level of AQP2and AQP3showed no significant difference between three groups. AQP4was expressed on the squamous epithelium, the pseudo multiple cilium columnar epithelium, the glands of the lamina propria mucosa and the muscular cells. The expression level of AQP4of the control group was higher than that of the model group, the expression level of the model group was higher than that of the aged group. AQP5was expressed on the the pseudo multiple cilium columnar epithelium, basal cells, serous glands and pipes. The expression level of AQP5of the control group was lower than that of the aged group, the expression level of the aged group was lower than that of the model group.Conclusion:The content of HA in the larynx of the D-galactose induced aging guinea pig was lower than that of the young on certain degree, while the content of the collagen was higher. Maybe the D-galactose induced aging guinea pig can be used for presbyphonia. AQP1, AQP2, AQP3, AQP4, AQP5were expressed in the larynx of the guinea pig, maybe AQPs involve in the aging course. AQP1, AQP4and AQP5may be associated with the aging of the guinea pig larynx.
