Topiramate is a newly-developed antiepileptic drug, whose structure is totally different to tranditional drugs. Though marketed domestically and abroad, the antiepileptic mechanism of topiramate is still unknown, and the researches on animal experiment have not been reported neither. We investigated the antiepileptic effect and the mechanism of topiramate on amygdala kindling in rats. The results are as follows:1 Amygdala kindling models were established successfully.2 Topiramate 50-200 mg/kg ig dose-dependently inhibited amygdala kindled seizure in rats, shortened the afterdischarge duration (ADD) and reduced the Racine's stage.(P<0.01)3 The combination of ineffective doses of topiramate 50 mg/kg ig and valproate sodium 200 mg/kg ig shortened ADD and reduced Racine's stage.( P<0.05)4 The combination of ineffective doses of topiramate 50 mg/kg ig and nicardipine 2 mg/kg ip shortened ADD. ( P<0.05)5 Topiramate 200 mg/kg ig significantly reduced the rate of seizure and death of semicarbazide-induced convulsion in mice. (P<0.01)The results suggested that topiramate inhibit the seizure of the amygdalakindling. Its mechanism is probably related to enchance the GABAergic system and block the calcium channel, which provide a new evidence to treatepilepsy reasonably with antiepileptics, especially in combined situationduring clinical treatment on epilepsy.
