| Objective To investigate the effect of Puerarin on platelet activation after balloon angioplasty.Methods 63 Wister rats were randomly devided into 3 groups rGroup 1 (n=21) served as controls. Group 2 (n=21) were given vascular balloon injury. Group 3 (n=21) were given Puerarin (50mg/kg/d ip.) from 1 day before injury to 14 days after injury. The level of plasma TXB2 and 6-K-PGFia and the number of alpha-granule membrane glycoprotein ?40 (GMP-140) molecule on platelet surface were measured at day 3 , 7 and 14 after balloon injury. Results (1) The number of platelet membrane GMP-140 of group 2 were obviously higher than group 1 after balloon injury at day 3 ( PO.01 );The number began to go down after 1 week (PO.05 ) and reach the same level as group 1 till 2 weeks. The number of platelet membrane GMP-140 of group 3 were lower than that of group 2 at every time (PO.01, PO.01, P> 0.05 ) .There were no difference between group 3 and group 1 (P>0.05, P>0.05, P>0.05 ). (2) The level of group 2 of plasma TXB2 were higher than group 1 at day 3 and 7 after balloon injury (PO.05 ). There were no difference between group 2 and group 1 ( P>0.05 ). The level of plasma TXB2 of group 3 were lower than group 2 at every time (PO.05, PO.05, P>0.05 ) and had no difference with group 1 ( P>0.05, P>0.05, P>0.05 ). (3) The level of plasma 6-Keto-PGFi a of group 2 were lower than that of group 1 .The difference were obviously at day 3 and 7 (PO.01, PO.01 ) and there were no difference till day 14 (P>0.05 ).The level of plasma 6-Keto-PGFi a of group 3 were obviously higher than group 2,especially at day 3 and 7 (PO.01, PO.01 ). There were no difference between group 2 and group 3 after 2 weeks ( P>0.05, P>0.05 ). Conclution The platelets are at the state of activation at least 1 week after balloon injury. Puerarin can lower platelet activation and decrease the adhesion between platelets and intimal. Puerarin contribute to antithrombotic and avoiding thrombotion to a certain extent.
