Lung carcinoma is a common kind of malignant tumors which menace human health, the prognosis is poor ,the mechanism and oncogenesis is not clear. Burnet developed the immunosurveillance concept in 1971. Malignant cells obviously escape immune surveillance, which suggests that they possess the ability of evading any potentially lethal effector mechanisms operated by the immune system. Potential strategies that tumors may use to avoid host immune responses include down-regulation of major histocompatibility complex molecules, expressing of poorly immunogenic tumor antigens, masking or shedding of immunogenic tumor antigens, induction of tolerance via display of an incomplete antigen-presention function, and secretion of immuno- suppressive factors. The status of immune system significanlty correlated with the development and clinicalcourse of lung cancer.Recently, many studies have focused on the cytokine network involvedin the tumor microenviroment. In the analysis of the cytokine network, Tlymphocytes have been found to express two distinct cytokine patterns: type1 cytokines, which include interleukin(IL)-2 and interferon- Y and havebeen demonstrated to promote cell-mediated immunity, and type 2 cytokines, which include IL-4,IL-5,IL-6,IL-10 and IL-13 and have been shown tosuppress cellular immune responses. Thl and Th2 cells oppose oneanothers' activity, cytokine production and differentiation.Thepredominance of the type 2 cytokine pattern is reported in cancer patientsrecently. It is one of the mechanisms of escaping from immune surveillance.IL-2 was identified as a key cytokine secreted by Thl-T-cells. Some studiesdemonstrates in a large series of patients that in advanced NSCLC there isan imbalance of the IL-2/IL-2 receptor system. Furthermore, circulatingSIL-2R levels and the SIL-2R/IL-2 ratio may be useful as markers ofdisease activity and treatment response, suggesting a potential prognosticvalue. IL-10 was identified as a major cytokine secreted by Th2-T-cells, itpossesses several inhibitory effects on the generation of anti-tumorimmunity. In patients with NSCLC, increased production ofimmunosuppressive IL-10 by lung cancer cells and increased plasma IL-10concentrations have recently been correlated to reduced survival. Type 2cytokine expression is predominant in the malignant disease and these cytokines may mediate immunosuppression. The measure that alter this type 2 predominant pattern of typel/type2 axis may represent promising future strategies of oncological therapy. In this study, to evaluate the correlation between the lung cancer and the level of serum IL-1CK SIL-2R and IL-2, we measured the IL-KK SIL-2R and IL-2 serum levels of 71 lung cancer. It may not only be involved in the development and course of lung cancer but also serve as prognostic markers for survival and, perhaps even more important, as predictive markers for the efficacy of a specific therapeutic strategy.1. Patients and Methods1.1 Patients1.1.1 The study population consisted 71 previously untreated patients with lung cancer. Among them, 21 patients had squamous cell carcinoma. 21 adenocarcinoma, 10 small cell carcinoma and 17 others. Forty nine patients are male and 22 patients are female. Mean age 60.4 years, range 24-86 years.1.1.2 The control population consisted of 30 benign lung disease, it contained pneumonias, tuberculous pleural effusion and others. Twenty-four patients are male and 6 patients are female. Mean age56.3 years, range 27-83 years. 1.1.3 10 healthy contro 1 subj ects.1.2 Sampling and Storage: Blood samples were obtained from patients before treatment and stored at -20癈 until assayed for cytokines.1.3 Methods: Cytikines analysis was performed using the enzyme-linked-immunosorbent-assay(ELISA) methods.1.4 Statistical analysis: The serum cytokine levels were presented as means and standard deviations.
