Objective: To investigate the antigenic specificity of a new hepatocellular carcinoma (HCC) associated antigen Kinectin in HCC.Methods: Recombinant Kinectin antigen was expressed in Ecoli with transfection of X phage. Following blotting the protein to nitrocellulose membrane and immunocytochemistry were used to detect anti-Kinectin antibody in a panel of HCC patients' sera and the sera from healthy individuals.Result: Of thirty-two sera from HCC patients, there was twentyshowed antibody-positive responses to Kinectin (20/32, 62.5%) ; and of fifty sera from healthy individuals , there was three showed antibody-positive responses to Kinectin (3/50, 6%), which had significance between them (p<0.01). Furthermore, there were no distinct differences among anti-Kinectin antibody, AFP, the tumor size, pathological grade and ages. The sensitivity, specificity and accuracy of Kinectin in the diagnosis of HCC were 62.5%, 94% and 81.7%, respectively.Conclusion: With certain specificity in HCC , Kinectin might have some clinic significance and be considered as one of new serological marker in diagnosis of HCC.
