Objective:To investigate the effect that the human B lymphocyte stimulated by kinectin protein induces the effector T lymphocyte to kill hepatocellular carcinoma cells.Method:Firstly, MBP-kinectin fusion protein was obtained by inducing Rosetta bacteria with pMAL-C2/kinectin 1-8a plasmid. Secondly, mononuclearcell was separateed from the healthal human peripheral blood and then cultured in vitro. During cultivation, B cell activating factor (BAFF) and MBP-kinectin were added. After being disassociated with the method of nylon wool column from mononuclearcell cultured in vitro, T lymphocyte was incubated together with hepatocellular carcinoma cell strain 7404. The effect of T lymphocyte killing hepatoma carcinoma cells was detected by LDH release assay.Result:MBP-kinectin fusion protein was obtained. Killing ratio of T lymphocyte for killing hepatoma carcinoma cells was as follows:control group was 3.40%±0.27%; experimental group was 38.48%±2.64%; negative-control group was 13.03%±2.38%. The data were analyzed by statistical software and showed the significant difference among 3 groups in statistics.Conclusion:Human B lymphocyte stimulated by kinectin fusion protein was able to induce effector T lymphocyte killing hepatoma carcinoma cell strain 7404.
