Objective:Based on the previous clinical and experimental results, this research deals with the mechanism that acupuncture therapy of "Tiaoshenyizhi" suppressed apoptosis in brain from genes relative to supply new experimental proofs for MID treated with acupuncture. Method:The rat MID model was rnade by injecting thrombi into the internal carotid artery. The rats were randomly divided into model group acupuncture group . non-acupoint group and pseudo-operating group by their weight. The stimulating points for acupuncture group were "Renzhong "Neiguan" and "Fengchi", those for non-acupcint group two fixed non-acupoints at hypochondria. Both of them were needled one time every day. Model group and pseudo-operating group were stimulaied by grasp at the same time and strength as acupuncture group and non-acupoint group. After two weeks, all rats were killed by cervical dislocation. The left brains of them were taken out. The expression of bcl-2 mRNA and its protein box mRNA and its protein in cortex hippocampus corpus striatum and thalamus were determined by in situ hybridization and immunocytochemical technique with computerized image processing. The datum were analyzed with one-way ANOVA by means of SPSS 10.0. Results:1 The expression of bcl-2 mRNA in cortex hippocampus corpus striatum and thalamus in model group were significantly higher than that in pseudo-operating group(P<0.05).The expression of bcl-2 mRNA in cortex hippocampus corpus striatum and thalamus in acupuncture group were significantly higher than that in model group and non-acupoint group(P<0.05).2 The expression of bcl-2 protein in cortex corpus striatum and thalamus in model group were significantly higher than that in pseudo-operating group(P<0.05).Although the expression of bcl-2 protein in hippocampus in model group had a tendency to enhance, there was no significant difference(P>)0.05 between two groups. The expression of bcl-2 protein in cortex hippocampus corpus striatum and thalamus in acupuncture group were significantly higher than that in model group and non-acupoint group(P<0.05).3 The expression of bax mRNA and its protein in cortex hippocampuscorpus striatum and thalamus in model group were significantly higher thanthat in pseudo-operating group(P<0.05).The expression of bax mRNA and itsprotein in cortex . hippocampus corpus striatum and thalamus inacupuncture group were significantly higher than that in model group andnon-acupoint group(P<0.05).4 The ratio of bcl-2 to bax in acupuncture group were higher than that inmodel group.Conclusion:1 Acupuncture can enhance the expression of bcl-2 supress the expression ofbax and improve the ratio of bcl-2 to bax in the brain of rat MID model. Thismay be one of mechanisms for acupuncture to supress aooptosis and protectcells of brain.2 There is a special property for acupuncture to regulate the expression ofbcl-2 and bax in brain of rat MID model.
